Purification and immunochemical studies of pyruvate dehydrogenase complex from rat heart, and cell-free synthesis of lipoamide dehydrogenase, a component of the complex.

TLDR
In cell-free translation programmed with free and membrane-bound polysomes, activity of mRNA coding for the precursor of the enzyme was much higher in freepolysomes than in membrane- bound polysome, and its translation product was detected as a putative precursor larger than the mature subunit.

Regulation of mitochondrial pyruvate dehydrogenase activity by tau protein kinase I/glycogen synthase kinase 3beta in brain

TL;DR: Results suggest that TPKI/GSK-3beta plays a key role in the pathogenesis of Alzheimer disease and regulates PDH and participates in energy metabolism and acetylcholine synthesis.

Autoantibodies of primary biliary cirrhosis recognize dihydrolipoamide acetyltransferase and inhibit enzyme function.

TL;DR: Identification of dihydrolipoamide acetyltransferase as the target of autoimmunity in PBC provides a reagent that can be used to determine mechanisms by which this molecule is recognized, and will allow study of whether autoimmune reactivity, at the humoral or T cell level, is the basis for the pathogenesis of PBC.

Dihydrolipoamide dehydrogenase: Functional similarities and divergent evolution of the pyridine nucleotide-disulfide oxidoreductases

TL;DR: The comparison of the complete amino acid sequences of E3 from Escherichia coli, yeast, pig, and human shows considerable homologies of certain amino acid residues or short stretches of sequences, especially in the specific catalytic and structural domains.

Isolation and sequence determination of cDNA clones for porcine and human lipoamide dehydrogenase: homology to other disulfide oxidoreductases

TL;DR: Alignment of the deduced amino acid sequence of human lipoamide dehydrogenase with human erythrocyte glutathione reductase and mercuric reduct enzyme from Tn501 revealed extensive homologies throughout the primary sequence, suggesting that secondary and tertiary structure is also similar among these three enzymes.
Journal ArticleDOI

Cloning and cDNA sequence of the dihydrolipoamide dehydrogenase component human alpha-ketoacid dehydrogenase complexes.

TL;DR: By blot-hybridization analysis with the cDNA as probe, two mRNAs, 2.2 and 2.4 kilobases in size, have been detected in human tissues and fibroblasts, whereas only one mRNA was detected in rat tissues.
