Journal ArticleDOI
Single-cell mutation analysis of clonal evolution in myeloid malignancies.
Linde A. Miles,Robert L. Bowman,Tiffany R. Merlinsky,Isabelle S. Csete,Aik Ooi,Robert Durruthy-Durruthy,Michael Bowman,Christopher Famulare,Minal Patel,Pedro Mendez,Chrysanthi Ainali,Benjamin Demaree,Cyrille L. Delley,Adam R. Abate,Manimozhi Manivannan,Sombeet Sahu,Aaron D Goldberg,Kelly L. Bolton,Ahmet Zehir,Raajit K. Rampal,Martin Carroll,Sara E. Meyer,Aaron D. Viny,Ross L. Levine +23 more
TL;DR: Single cell clonal architecture provides novel insights into the pathogenesis of myeloid transformation and how clonal complexity evolves with disease progression and is combined with mutational analysis to map somatic genotype and clonal Architecture with immunophenotype.
Journal ArticleDOI

Bagnoli,Wolfgang Enard,Marta Gut,Rickard Sandberg,Itoshi Nikaido,Ivo Gut,Oliver Stegle,Oliver Stegle,Holger Heyn +43 more
TL;DR: In this paper, a multicenter study comparing 13 commonly used scRNA-seq and single-nucleus RNA-seq protocols applied to a heterogeneous reference sample resource was conducted.
Journal ArticleDOI
Novel Stem/Progenitor Cell Population from Murine Tracheal Submucosal Gland Ducts with Multipotent Regenerative Potential
Ahmed E. Hegab,Vi Luan Ha,Jennifer L. Gilbert,Kelvin Xi Zhang,Stephen P. Malkoski,Andy T. Chon,Daphne O. Darmawan,Bharti Bisht,Aik Ooi,Matteo Pellegrini,Derek W. Nickerson,Brigitte N. Gomperts +11 more
TL;DR: It is demonstrated that airway submucosal gland (SMG) duct cells, in addition to BCs, survived severe hypoxic‐ischemic injury and are therefore a multipotent stem cell for airway epithelial repair.
Journal ArticleDOI
Direct detection of double-stranded DNA: molecular methods and applications for DNA diagnostics
TL;DR: Progress is described in adapting polyamides, triplex DNA, and engineered zinc finger DNA-binding proteins as dsDNA diagnostic systems and the sequence-enabled reassembly (SEER) method, involving the use of custom zinc finger proteins, offers the potential for direct detection of double-stranded DNA in cells.
Journal ArticleDOI
DNA Sequence-Enabled Reassembly of the Green Fluorescent Protein
TL;DR: The feasibility of the SEER approach utilizing the split green fluorescent protein appended to appropriate zinc fingers, such that chromophore formation is only catalyzed in the presence of DNA sequences that incorporate binding sites for both zinc fingers.
