Journal ArticleDOI
Minimal functional driver gene heterogeneity among untreated metastases
Johannes G. Reiter,Johannes G. Reiter,Alvin Makohon-Moore,Jeffrey M. Gerold,Alexander Heyde,Marc A. Attiyeh,Zachary A. Kohutek,Collin Tokheim,Alexia Brown,Rayne M. DeBlasio,Juliana Niyazov,Amanda Zucker,Rachel Karchin,Rachel Karchin,Kenneth W. Kinzler,Christine A. Iacobuzio-Donahue,Bert Vogelstein,Martin A. Nowak +17 more
TL;DR: Within individual patients, a large majority of driver gene mutations are common to all metastases, and a mathematical model of tumor evolution and metastasis formation provides an explanation for the observed driver gene homogeneity.
Journal ArticleDOI
Increased stem cell proliferation in atherosclerosis accelerates clonal hematopoiesis
Alexander Heyde,David Rohde,Cameron S. McAlpine,Shuang Zhang,Friedrich Felix Hoyer,Jeffrey M. Gerold,David Cheek,Yoshiko Iwamoto,Maximilian J. Schloss,Katrien Vandoorne,Oriol Iborra-Egea,Christian Muñoz-Guijosa,Antoni Bayes-Genis,Johannes G. Reiter,Morgan Craig,Filip K. Swirski,Matthias Nahrendorf,Martin A. Nowak,Kamila Naxerova +18 more
TL;DR: In this paper, the authors show that hematopoietic stem cell division rates are increased in mice and humans with atherosclerosis, and that increased stem cell proliferation expedites somatic evolution and expansion of clones with driver mutations.
Journal ArticleDOI
The Evolutionary Origins of Recurrent Pancreatic Cancer.
Hitomi Sakamoto,Marc A. Attiyeh,Jeffrey M. Gerold,Alvin Makohon-Moore,Akimasa Hayashi,Jungeui Hong,Rajya Kappagantula,Lance Zhang,Jerry P. Melchor,Johannes G. Reiter,Alexander Heyde,Craig M. Bielski,Alexander V Penson,Mithat Gonen,Debyani Chakravarty,Eileen M. O'Reilly,Laura D. Wood,Ralph H. Hruban,Martin A. Nowak,Nicholas D. Socci,Barry S. Taylor,Christine A. Iacobuzio-Donahue +21 more
TL;DR: Routine post-treatment sampling may have value in the management of recurrent pancreatic cancer, and it is identified that recurrent disease after adjuvant or first-line platinum therapy corresponds to an increased mutational burden.
Journal ArticleDOI
Evolution of delayed resistance to immunotherapy in a melanoma responder
David Liu,David Liu,Jia-Ren Lin,Emily J. Robitschek,Emily J. Robitschek,Gyulnara G. Kasumova,Alexander Heyde,Alvin Shi,Alvin Shi,Adam A. Kraya,Gao Zhang,Gao Zhang,Tabea Moll,Dennie T. Frederick,Yu-An Chen,Shu Wang,Denis Schapiro,Denis Schapiro,Li Lun Ho,Li Lun Ho,Kevin Bi,Kevin Bi,Avinash Das Sahu,Shaolin Mei,Benchun Miao,Tatyana Sharova,Christopher Alvarez-Breckenridge,Jackson Stocking,Tommy Kim,Riley Fadden,Donald P. Lawrence,Mai P. Hoang,Daniel P. Cahill,Mohsen Malehmir,Mohsen Malehmir,Martin A. Nowak,Priscilla K. Brastianos,Christine G. Lian,Eytan Ruppin,Benjamin Izar,Meenhard Herlyn,Eliezer M. Van Allen,Eliezer M. Van Allen,Katherine L. Nathanson,Keith T. Flaherty,Ryan J. Sullivan,Manolis Kellis,Manolis Kellis,Peter K. Sorger,Genevieve M. Boland,Genevieve M. Boland +50 more
TL;DR: In this article, a high-resolution map of the evolutionary dynamics of resistance to ICB, characterizes a de-differentiated neural-crest tumor population in melanoma immunotherapy resistance and describes site-specific differences in tumor-immune interactions via longitudinal analysis of a patient with melanoma with an unusual clinical course.
Journal ArticleDOI
Consecutive seeding and transfer of genetic diversity in metastasis.
TL;DR: A mathematical model of consecutive seeding by multiple cells that relates the expected diversity of a metastasis to the diversity in the pool of seeding cells is derived and the probability that a metastases is polyclonal is calculated.
