Journal ArticleDOI
RNA Demethylase ALKBH5 Selectively Promotes Tumorigenesis and Cancer Stem Cell Self-Renewal in Acute Myeloid Leukemia.
Chao Shen,Yue Sheng,Allen C. Zhu,Sean Robinson,Xi Jiang,Xi Jiang,Lei Dong,Huiying Chen,Rui Su,Zhe Yin,Wei Li,Xiaolan Deng,Yinhuai Chen,Yinhuai Chen,Yueh-Chiang Hu,Yueh-Chiang Hu,Hengyou Weng,Huilin Huang,Emily Prince,Christopher R. Cogle,Miao Sun,Miao Sun,Bin Zhang,Chun-Wei Chen,Guido Marcucci,Chuan He,Zhijian Qian,Jianjun Chen +27 more
TL;DR: It is demonstrated that ALKBH5 is required for the development and maintenance of AML and self-renewal of leukemia stem/initiating cells (LSCs/LICs) but not essential for normal hematopoiesis, and the therapeutic potential of targeting the AL KBH5/m6A axis is highlighted.
Journal ArticleDOI
N6-Deoxyadenosine Methylation in Mammalian Mitochondrial DNA
Ziyang Hao,Tong Wu,Xiaolong Cui,Pingping Zhu,Cai-Ping Tan,Xiaoyang Dou,Kai-Wen Hsu,Yueh-Te Lin,Pei-Hua Peng,Li-Sheng Zhang,Yawei Gao,Lulu Hu,Hui-Lung Sun,Allen C. Zhu,Jianzhao Liu,Kou-Juey Wu,Chuan He +16 more
TL;DR: It is reported that mammalian mitochondrial DNA (mtDNA) is enriched for 6mA, which is at least 1,300-fold higher than that in gDNA under normal growth conditions, corresponding to approximately four 6mA modifications on each mtDNA molecule.
Journal ArticleDOI
Stabilization of ERK-Phosphorylated METTL3 by USP5 Increases m6A Methylation.
Hui-Lung Sun,Allen C. Zhu,Yawei Gao,Hideki Terajima,Hideki Terajima,Qili Fei,Qili Fei,Shun Liu,Shun Liu,Linda Zhang,Linda Zhang,Zijie Zhang,Zijie Zhang,Bryan T. Harada,Bryan T. Harada,Yu-Ying He,Marc Bissonnette,Mien Chie Hung,Chuan He,Chuan He +19 more
TL;DR: This study reveals an unrecognized function of ERK in regulating m6A methylation, found that ERK phosphorylates METTL3 at S43/S50/S525 and WTAP at S306/S341, followed by deubiquitination by USP5, resulting in stabilization of the m 6A methyltransferase complex.
Journal ArticleDOI
METTL3 Regulates Liver Homeostasis, Hepatocyte Ploidy, and Circadian Rhythm-Controlled Gene Expression in Mice.
Juan M. Barajas,Cho-Hao Lin,Hui-Lung Sun,Frances Alencastro,Allen C. Zhu,Mona Aljuhani,Ladan Navari,Selen A. Yilmaz,Lianbo Yu,Kara N. Corps,Chuan He,Andrew W. Duncan,Kalpana Ghoshal +12 more
TL;DR: In this paper, the role of METTL3 in liver biology was unraveled in mice depleted of Mettl3 in neonatal livers (Mettl 3fl/fl; Alb-Cre, "M3LKO").
