Pluripotent state transitions coordinate morphogenesis in mouse and human embryos
Marta N. Shahbazi,Antonio Scialdone,Natalia Skorupska,Antonia Weberling,Gaëlle Recher,Meng Zhu,Agnieszka Jedrusik,Liani Devito,Laila Noli,Iain C. Macaulay,Christa Buecker,Y. Khalaf,Dusko Ilic,Thierry Voet,Thierry Voet,John C. Marioni,John C. Marioni,John C. Marioni,Magdalena Zernicka-Goetz +18 more
TL;DR: It is shown that exit from an unrestricted naive pluripotent state is required for epiblast epithelialization and generation of the pro-amniotic cavity in mouse embryos and the relevance of transitions between these states during development of the mammalian embryo is shown.
Journal ArticleDOI
Morphogenesis of extra-embryonic tissues directs the remodelling of the mouse embryo at implantation.
Neophytos Christodoulou,Antonia Weberling,Douglas Strathdee,Kurt I. Anderson,Paul Timpson,Paul Timpson,Magdalena Zernicka-Goetz +6 more
TL;DR: The authors use live imaging with a cdx2-GFP reporter line in combination with loss of function experiments to demonstrate that FGF signalling mediated trophectoderm morphogenesis orchestrates this process.
Book ChapterDOI
Comparative analysis of human and mouse development: From zygote to pre-gastrulation
TL;DR: Current knowledge of mammalian development of both mouse and human is reviewed, focusing on morphogenetic processes leading to the onset of gastrulation, when the embryonic anterior-posterior axis becomes established and the three germ layers start to be specified.
Journal ArticleDOI
A single cell characterisation of human embryogenesis identifies pluripotency transitions and putative anterior hypoblast centre.
Matteo A. Molè,Matteo A. Molè,Tim H. H. Coorens,Marta N. Shahbazi,Marta N. Shahbazi,Antonia Weberling,Bailey A. T. Weatherbee,Carlos W. Gantner,Carmen Sancho-Serra,Lucy Richardson,Abbie Drinkwater,Najma Syed,Stephanie Engley,Philip Snell,Leila Christie,Kay Elder,Alison Campbell,Simon Fishel,Simon Fishel,Sam Behjati,Sam Behjati,Roser Vento-Tormo,Magdalena Zernicka-Goetz,Magdalena Zernicka-Goetz +23 more
TL;DR: In this article, the authors identify a group of asymmetrically positioned extra-embryonic hypoblast cells expressing inhibitors of BMP, NODAL and WNT signalling pathways and suggest that these cells can act as the anterior singalling centre to pattern the epiblast.
Journal ArticleDOI
Integrin-Mediated Focal Anchorage Drives Epithelial Zippering during Mouse Neural Tube Closure.
Matteo A. Molè,Matteo A. Molè,Gabriel L. Galea,Ana Rolo,Antonia Weberling,Oleksandr Nychyk,Oleksandr Nychyk,Sandra C. P. De Castro,Dawn Savery,Reinhard Fässler,Patricia Ybot-Gonzalez,Nicholas D. E. Greene,Andrew J. Copp +12 more
TL;DR: The molecular mechanism underlying zippering during mouse spinal neural tube closure is uncovered and integrin-mediated anchorage is proposed as an evolutionarily conserved mechanism of general relevance for zippered closure of epithelial gaps whose disturbance can produce clinically important birth defects.
