Journal ArticleDOI
Selective autophagy as a potential therapeutic target for neurodegenerative disorders.
TL;DR: A change of strategy in the modulation of autophagy might hold promise for future disease-modifying therapies for patients with neurodegenerative disorders.
Journal ArticleDOI
Chaperone-mediated autophagy prevents collapse of the neuronal metastable proteome.
Mathieu Bourdenx,Adrián Martín-Segura,Aurora Scrivo,Jose A. Rodriguez-Navarro,Susmita Kaushik,Inmaculada Tasset,Antonio Diaz,Nadia J. Storm,Qisheng Xin,Yves R. Juste,Erica Stevenson,Enrique Luengo,Cristina C. Clement,Se Joon Choi,Nevan J. Krogan,Nevan J. Krogan,Eugene V. Mosharov,Laura Santambrogio,Fiona Grueninger,Ludovic Collin,Danielle L. Swaney,Danielle L. Swaney,David Sulzer,Evripidis Gavathiotis,Ana Maria Cuervo +24 more
TL;DR: In this article, the role of chaperone-mediated autophagy (CMA) in neuronal proteostasis was investigated using mouse models with systemic and neuronal-specific CMA blockage, showing that loss of neuronal CMA leads to altered neuronal function, selective changes in the neuronal metastable proteome, and proteotoxicity, all reminiscent of brain aging.
Journal ArticleDOI
The different autophagy degradation pathways and neurodegeneration
Angeleen Fleming,Mathieu Bourdenx,M Fujimaki,Cansu Karabiyik,Gregory J. Krause,Ana Lopez,Adrián Martín-Segura,Claudia Puri,Aurora Scrivo,John Skidmore,Sung Min Son,Eleanna Stamatakou,Lidia Wrobel,Ye Zhu,Ana Maria Cuervo,David C. Rubinsztein +15 more
TL;DR: The term autophagy encompasses different pathways that route cytoplasmic material to lysosomes for degradation and includes macroautophagy, chaperone-mediated autophathy, and microautophathy as discussed by the authors .
Journal ArticleDOI
Monitoring spatiotemporal changes in chaperone-mediated autophagy in vivo.
Shuxian Dong,C. Aguirre-Hernandez,Aurora Scrivo,Carolina Eliscovich,Esperanza Arias,Jose Javier Bravo-Cordero,Ana Maria Cuervo,Ana Maria Cuervo +7 more
TL;DR: A transgenic reporter mouse is developed that allows dynamic measurement of CMA activity in vivo using image-based procedures, and previously unknown spatial and temporal differences in CMAActivity in multiple organs and in response to stress are identified.
Journal ArticleDOI
TSC1 loss increases risk for tauopathy by inducing tau acetylation and preventing tau clearance via chaperone-mediated autophagy
Carolina Alquezar,Kathleen M. Schoch,Ethan G. Geier,Eliana Marisa Ramos,Aurora Scrivo,Kathy H. Li,Andrea R. Argouarch,Elisabeth E. Mlynarski,Beth A. Dombroski,Michael DeTure,Dennis W. Dickson,Jennifer S. Yokoyama,Ana Maria Cuervo,Alma L. Burlingame,Gerard D. Schellenberg,Timothy M. Miller,Bruce L. Miller,Aimee W. Kao +17 more
TL;DR: In this article, a loss-of-function mutation in the TSC1 gene was linked to tau accumulative disorders, including age-associated neurodegenerative disorders demonstrating tau-laden intracellular inclusions.
