Lopez,Cécile K. Lopez,Isabelle Plo,Isabelle Plo,Isabelle Plo,Orianne Wagner-Ballon,Marcio Do Cruzeiro,François Delhommeau,François Delhommeau,Bertrand Arnulf,Marc-Henri Stern,Lucy A. TL;DR: It is reported that inactivation of Tet2 in mouse perturbs both early and late steps of hematopoiesis including myeloid and lymphoid differentiation in a cell-autonomous manner, endows the cells with competitive advantage, and eventually leads to the development of malignancies.
Journal ArticleDOI
Identification of Regulators of Polyploidization Presents Therapeutic Targets for Treatment of AMKL
Qiang Wen,Benjamin Goldenson,Serena J. Silver,Monica Schenone,Vlado Dančík,Zan Huang,Lingzhi Wang,Timothy A. Lewis,W. Carr,David E. Root,Stuart L. Schreiber,Andrew M. Stern,John D. Crispino +48 more
TL;DR: In this article, a high-content image-based screen was used to identify small-molecule probes that induce polyploidization of megakaryocytic leukemia cells and serve as perturbagens to help understand this process.
Journal ArticleDOI
Characterization of novel genomic alterations and therapeutic approaches using acute megakaryoblastic leukemia xenograft models
Clarisse Thiollier,Clarisse Thiollier,Clarisse Thiollier,Cécile K. Lopez,Cécile K. Lopez,Bastien Gerby,Cathy Ignacimouttou,Cathy Ignacimouttou,Sandrine Poglio,Yannis Duffourd,Justine Guegan,Paola Rivera-Munoz,Paola Rivera-Munoz,Paola Rivera-Munoz,Olivier Bluteau,Olivier Bluteau,Vinciane Mabialah,Vinciane Mabialah,Vinciane Mabialah,M'Boyba Diop,Qiang Wen,Arnaud Petit,Anne Laure Bauchet,Dirk Reinhardt,Beat Bornhauser,Daniel Gautheret,Daniel Gautheret,Yann Lécluse,Judith Landman-Parker,Isabelle Radford,William Vainchenker,William Vainchenker,Nicole Dastugue,Stéphane de Botton,Philippe Dessen,Philippe Dessen,Philippe Dessen,Jean-Pierre Bourquin,John D. Crispino,Paola Ballerini,Olivier Bernard,Olivier Bernard,Olivier Bernard,Françoise Pflumio,Thomas Mercher,Thomas Mercher,Thomas Mercher +46 more
TL;DR: A CBFA2T3-GLIS2 fusion gene was identified in 31% of non–Down syndrome AMKL, and it is unclear whether this gene is related to Down syndrome or not.
Journal ArticleDOI
Crosstalk between Notch and AKT signaling during murine megakaryocyte lineage specification
Melanie G. Cornejo,Vinciane Mabialah,Vinciane Mabialah,Stephen M. Sykes,Tulasi Khandan,Cristina Lo Celso,Cécile K. Lopez,Cécile K. Lopez,Paola Rivera-Munoz,Paola Rivera-Munoz,Philippe Rameau,Zuzana Tothova,Jon C. Aster,Ronald A. DePinho,David T. Scadden,D. Gary Gilliland,Thomas Mercher,Thomas Mercher,Thomas Mercher +18 more
TL;DR: The role of downstream mediators of NOTCH during megakaryopoiesis are investigated and crosstalk between the NOTCH and PI3K/AKT pathways is reported and the inhibitory role of phosphatase with tensin homolog and Forkhead Box class O factors on megakarianocyte lineage specification in vivo is demonstrated.
Journal ArticleDOI
DNMT3A(R882H) mutant and Tet2 inactivation cooperate in the deregulation of DNA methylation control to induce lymphoid malignancies in mice.
Laurianne Scourzic,Lucile Couronné,Marianne Terndrup Pedersen,V Della Valle,M'Boyba Diop,Elena Mylonas,Julien Calvo,Enguerran Mouly,Cécile K. Lopez,Cécile K. Lopez,Cécile K. Lopez,N Martin,Michaela Fontenay,Ambre Bender,Sylvain Guibert,Patrice Dubreuil,Philippe Dessen,Nathalie Droin,Françoise Pflumio,Michael Weber,P. Gaulard,P. Gaulard,Kristian Helin,Thomas Mercher,Thomas Mercher,Thomas Mercher,O. Bernard +26 more
TL;DR: The data confirm the transformation potential of DNMT3AR882H Tet2−/− progenitors and represent the first cooperative model in mice involving Tet2 inactivation driving lymphoid malignancies.
