Journal ArticleDOI
A map of open chromatin in human pancreatic islets
Kyle J. Gaulton,Takao Nammo,Lorenzo Pasquali,Jeremy M. Simon,Paul G. Giresi,Marie P. Fogarty,Tami M. Panhuis,Piotr A. Mieczkowski,Antonio Secchi,Domenico Bosco,Thierry Berney,Eduard Montanya,Karen L. Mohlke,Jason D. Lieb,Jorge Ferrer +14 more
TL;DR: It is found that human islet samples heterozygous for rs7903146 showed allelic imbalance in islet FAIRE signals and that the variant alters enhancer activity, indicating that genetic variation at this locus acts in cis with local chromatin and regulatory changes.
Journal ArticleDOI
Unique arrangement of alpha- and beta-cells in human islets of Langerhans
Domenico Bosco,Mathieu Pierre Jean Armanet,Philippe Morel,Nadja Niclauss,Antonino Sgroi,Yannick D. Muller,Laurianne Giovannoni,Géraldine Parnaud,Thierry Berney +8 more
TL;DR: Human islets have a unique architecture allowing all endocrine cells to be adjacent to blood vessels and favoring heterologous contacts between β- and α-cells, while permitting homologous contact between α- and β-cells.
Journal ArticleDOI
Beta Cell Hubs Dictate Pancreatic Islet Responses to Glucose
Natalie R. Johnston,Ryan K. Mitchell,Elizabeth Haythorne,Maria R. Paiva Pessoa,Francesca Semplici,Jorge Ferrer,Lorenzo Piemonti,Piero Marchetti,Marco Bugliani,Domenico Bosco,Ekaterine Berishvili,Philip Duncanson,Michael Watkinson,Johannes Broichhagen,Dirk Trauner,Guy A. Rutter,David J. Hodson,David J. Hodson,David J. Hodson +18 more
TL;DR: It is shown here that the β cell population in situ is operationally heterogeneous, and the islet is wired by hubs, whose failure may contribute to type 2 diabetes mellitus.
Journal ArticleDOI
Sulfonylurea induced beta-cell apoptosis in cultured human islets
Kathrin Maedler,Richard D. Carr,Domenico Bosco,Richard A. Zuellig,Thierry Berney,Marc Y. Donath +5 more
TL;DR: Closure of the inwardly rectifying K(+) sulfonylurea receptor subtype of ATP-sensitive potassium channels induces beta-cell apoptosis in human islets and may precipitate the decrease in beta- cell mass observed in patients with type 2 diabetes.
Journal ArticleDOI
Human bone marrow mesenchymal stem cells can express insulin and key transcription factors of the endocrine pancreas developmental pathway upon genetic and/or microenvironmental manipulation in vitro
C. Moriscot,Florence de Fraipont,Marie-Jeanne Richard,Mélanie Marchand,Mélanie Marchand,Pierre Savatier,Pierre Savatier,Domenico Bosco,Marie Favrot,Pierre‐Yves Benhamou +9 more
TL;DR: The results demonstrated that in vitro human bone marrow stem cells are able to differentiate into insulin‐expressing cells by a mechanism involving several transcription factors of the beta‐cell developmental pathway when cultured in an appropriate microenvironment.
