IL-6 triggers malignant features in mammospheres from human ductal breast carcinoma and normal mammary gland
Pasquale Sansone,Gianluca Storci,Simona Tavolari,Tiziana Guarnieri,Catia Giovannini,Mario Taffurelli,Claudio Ceccarelli,Donatella Santini,Paola Paterini,Kenneth B. Marcu,Pasquale Chieco,Massimiliano Bonafè +11 more
TL;DR: The hypothesis that IL-6 induces malignant features in Notch-3-expressing stem/progenitor cells from human ductal breast carcinoma and normal mammary gland is supported.
Journal ArticleDOI
The IL-6/JAK/Stat3 Feed-Forward Loop Drives Tumorigenesis and Metastasis
Qing Chang,Eirini Bournazou,Pasquale Sansone,Marjan Berishaj,Sizhi Paul Gao,Laura Daly,Jared Wels,Till Martin Theilen,Selena Granitto,Xinmin Zhang,Jesse W. Cotari,Mary L. Alpaugh,Elisa de Stanchina,Katia Manova,Ming O. Li,Massimiliano Bonafè,Claudio Ceccarelli,Mario Taffurelli,Donatella Santini,Grégoire Altan-Bonnet,Rosandra N. Kaplan,Larry Norton,Norihiro Nishimoto,Dennis Huszar,David Lyden,David Lyden,Jacqueline Bromberg +26 more
TL;DR: This study provides the first evidence for IL-6 expression at the leading edge of invasive human breast tumors and demonstrates mechanistically that IL- 6/JAK/Stat3 signaling plays a critical and pharmacologically targetable role in orchestrating the composition of the tumor microenvironment that promotes growth, invasion, and metastasis.
Journal ArticleDOI
Antigen retrieval techniques in immunohistochemistry: comparison of different methods.
Stefano Pileri,Giovanna Roncador,Claudio Ceccarelli,Milena Piccioli,Aspasia Briskomatis,Elena Sabattini,Stefano Ascani,Donatella Santini,Pier Paolo Piccaluga,Ornella Leone,Stefania Damiani,Cesarina Ercolessi,Federica Sandri,F. Pieri,Lorenzo Leoncini,Brunangelo Falini +15 more
TL;DR: Routine sections of normal and pathological samples fixed in 10 per cent buffered formalin or B5, including EDTA‐decalcified bone‐marrow biopsies, were tested with 61 antibodies following heating, and EDTA appeared to be superior in terms of both staining intensity and the number of marked cells.
Journal ArticleDOI
Mucosal immune activation in irritable bowel syndrome: gender-dependence and association with digestive symptoms
Cesare Cremon,Luciana Gargano,Antonio Maria Morselli-Labate,Donatella Santini,Rosanna Cogliandro,Roberto De Giorgio,Vincenzo Stanghellini,Roberto Corinaldesi,Giovanni Barbara +8 more
TL;DR: A large subset of IBS patients shows gender-dependent mucosal infiltration of immunocytes that correlates with abdominal bloating and dysmotility-like dyspepsia, and provides the rationale for considering immune mechanisms as a pathophysiological component in a subset ofIBS patients.
Journal ArticleDOI
Axillary dissection versus no axillary dissection in patients with breast cancer and sentinel-node micrometastases (IBCSG 23-01): 10-year follow-up of a randomised, controlled phase 3 trial.

TL;DR: The findings of the IBCSG 23-01 trial corroborate those obtained at 5 years and are consistent with those of the 10-year follow-up analysis of the Z0011 trial, and support the current practice of not doing an axillary dissection when the tumour burden in the sentinel nodes is minimal or moderate.
