Functional gamma-secretase inhibitors reduce beta-amyloid peptide levels in brain
H. F. Dovey,V. John,Chen Jin Ling,P. D. S. int Andrieu,L. Y. Fang,S. B. Freedman,B. Folmer,Erich Goldbach,E. J. Holsztynska,Kun Hu,K. L. Johnson Wood,S. L. Kennedy,Dora Kholodenko,J. E. Knops,L. H. Latimer,Michael K. Lee,Z. Liao,I. M. Lieberburg,Ruth Motter,L. C. Mutter,J. Nietz +20 more
Journal ArticleDOI
Reduction of beta-amyloid peptide42 in the cerebrospinal fluid of patients with Alzheimer's disease.
R. Motter,Carmen Vigo-Pelfrey,Dora Kholodenko,Robin Barbour,Kelly Johnson-Wood,Doug R. Galasko,Linda Chang,Bruce L. Miller,C. Clark,Robert C. Green,D. Olson,P. Southwick,R. Wolfert,B. Munroe,Ivan Lieberburg,Peter Seubert,Dale Schenk +16 more
TL;DR: It is interesting that CSF Aβ42 levels were found to be significantly lower in AD patients relative to controls, whereas total Aβ levels were not, and neither A β42 nor τ levels were apparently influenced by the ApoE genotype.
Journal ArticleDOI
Epitope and isotype specificities of antibodies to β-amyloid peptide for protection against Alzheimer's disease-like neuropathology
Frederique Bard,Robin Barbour,Catherine Cannon,Robert Carretto,Michael Fox,Dora Games,Teresa Guido,Kathleen Hoenow,Kang Hu,Kelly Johnson-Wood,Karen Khan,Dora Kholodenko,Celeste Lee,Michael K. Lee,Ruth Motter,Minh Nguyen,Amanda Reed,Dale Schenk,Pearl Tang,Vasquez Nicki J,P. Seubert,Ted Yednock +21 more
TL;DR: Results indicate that antibody Fc-mediated plaque clearance is a highly efficient and effective process for protection against neuropathology in an animal model of Alzheimer's disease.
Journal ArticleDOI
High Cerebrospinal Fluid Tau and Low Amyloid β42 Levels in the Clinical Diagnosis of Alzheimer Disease and Relation to Apolipoprotein E Genotype
Doug R. Galasko,Linda Chang,Ruth Motter,Christopher M. Clark,J. A. Kaye,David S. Knopman,Ronald G. Thomas,Dora Kholodenko,Dale Schenk,I. Lieberburg,Bruce L. Miller,Robert C. Green,R. Basherad,L. Kertiles,M. A. Boss,P. Seubert +15 more
TL;DR: Combined analysis of CSF Abeta42 and tau levels discriminated patients with AD, including patients with mild dementia, from the NC group, supporting use of these proteins to identify AD and to distinguish early AD from aging.
Journal ArticleDOI
Enhanced Production and Oligomerization of the 42-residue Amyloid β-Protein by Chinese Hamster Ovary Cells Stably Expressing Mutant Presenilins
Weiming Xia,Jimin Zhang,Dora Kholodenko,Martin Citron,Marcia B. Podlisny,David B. Teplow,Christian Haass,Peter Seubert,Edward H. Koo,Dennis J. Selkoe +9 more
TL;DR: It is concluded that mutant PS1 and PS2 proteins enhance the proteolysis of β-amyloid precursor protein by the γ-secretase cleaving at Aβ residue 42, thereby promoting amyloidogenesis.
