Journal ArticleDOI
Use of overlapping peptide mixtures as antigens for cytokine flow cytometry
Holden T. Maecker,Holli S Dunn,Maria A. Suni,Elham Khatamzas,Elham Khatamzas,Christine J. Pitcher,Torsten Bunde,Natasha Persaud,Wendy L. Trigona,Tong-Ming Fu,Elizabeth Sinclair,Barry M. Bredt,Joseph M. McCune,Vernon C. Maino,Florian Kern,Louis J. Picker +15 more
TL;DR: Over overlapping 15 amino acid peptide mixes may facilitate the analysis of antigen-specific CD4+ and CD8+ T-cell responses by cytokine flow cytometry, using clinical specimens that include shipped blood or cryopreserved PBMC.
Journal ArticleDOI
Cytomegalovirus (CMV) phosphoprotein 65 makes a large contribution to shaping the T cell repertoire in CMV-exposed individuals
Florian Kern,Torsten Bunde,Nicole Faulhaber,Nicole Faulhaber,Felix Kiecker,Elham Khatamzas,Ina Maria Rudawski,Axel Pruss,Jan W. Gratama,Rudolf Volkmer-Engert,Ralf Ewert,Petra Reinke,Hans-Dieter Volk,Louis J. Picker +13 more
TL;DR: Antigen-specific, cytokine flow cytometry was used to analyze the prevalence and frequency of CD4 and CD8 memory T cells specific for the abundantly expressed cytomegalovirus (CMV) phosphoprotein 65 in healthy CMV IgG-seropositive individuals and several new T cell epitopes were identified.
Journal ArticleDOI
Analysis of CD8 T cell reactivity to cytomegalovirus using protein-spanning pools of overlapping pentadecapeptides.
Florian Kern,Nicole Faulhaber,Claudia Frömmel,Elham Khatamzas,Susanna Prösch,Constanze Schönemann,Ines Kretzschmar,Rudolf Volkmer-Engert,Hans-Dieter Volk,Petra Reinke +9 more
TL;DR: The results confirmed the previous contention that HCMV IE‐1 is an important CD8 T cell target and the technical improvement made in order to address this question has clearly wider implications.
Journal ArticleDOI
Distribution of human CMV-specific memory T cells among the CD8pos. subsets defined by CD57, CD27, and CD45 isoforms.
Florian Kern,Elham Khatamzas,Ingolf Surel,Claudia Frömmel,Petra Reinke,Shar L. Waldrop,Louis J. Picker,Hans-Dieter Volk +7 more
TL;DR: A novel flow cytometric technique that allows simultaneous detection of human cytomegalovirus (HCMV)‐specific CD8pos.
Journal ArticleDOI
Clonotypic Structure of the Human CD4+ Memory T Cell Response to Cytomegalovirus
Arlene D. Bitmansour,Shar L. Waldrop,Christine J. Pitcher,Elham Khatamzas,Florian Kern,Vernon C. Maino,Louis J. Picker +6 more
TL;DR: The CMV-specific CD4+ T cell memory repertoire in normal subjects is characterized by striking clonotypic dominance and the potential for epitope focusing, suggesting that primary responsibility for immunosurveillance against CMV reactivation rests with a handful of clones recognizing a limited array of CMV determinants.
