A breast cancer stem cell niche supported by juxtacrine signalling from monocytes and macrophages
Haihui Lu,Karl R. Clauser,Wai Leong Tam,Julia Fröse,Xin Ye,Elinor Ng Eaton,Ferenc Reinhardt,Vera S. Donnenberg,Rohit Bhargava,Steven A. Carr,Robert A. Weinberg +10 more
TL;DR: It is shown that tumour-associated monocytes and macrophages create a CSC niche through juxtacrine signalling with CSCs through upregulating the expression of CD90 and EphA4, which mediate the physical interactions of C SCs with TAMs by directly binding with their respective counter-receptors on these cells.
Journal ArticleDOI
A Biochemically Defined System for Mammalian Nonhomologous DNA End Joining
Yunmei Ma,Haihui Lu,Brigette Tippin,Myron F. Goodman,Noriko Shimazaki,Osamu Koiwai,Chih-Lin Hsieh,Klaus Schwarz,Michael R. Lieber +8 more
TL;DR: The NHEJ reactions have been reconstituted in vitro by using purified Ku, DNA-PK(cs), Artemis, and XRCC4:DNA ligase IV proteins to join incompatible ends to yield diverse junctions to lead to an iterative strand-processing model for the steps of N HEJ.
Journal ArticleDOI
Protein Kinase C α Is a Central Signaling Node and Therapeutic Target for Breast Cancer Stem Cells
Wai Leong Tam,Haihui Lu,Joyce Y. Buikhuisen,Boon Seng Soh,Elgene Lim,Ferenc Reinhardt,Zhenhua Jeremy Wu,Jordan A. Krall,Brian Bierie,Wenjun Guo,Xi Chen,Xiaole Shirley Liu,Myles Brown,Bing Lim,Robert A. Weinberg +14 more
TL;DR: In this paper, an AP-1 molecular switch was identified in which c-FOS and FRA1 are preferentially utilized in non-CSCs and CSCs, respectively.
Journal ArticleDOI
Human Chromosomal Translocations at CpG Sites and a Theoretical Basis for their Lineage and Stage Specificity
Albert G. Tsai,Haihui Lu,Sathees C. Raghavan,Markus Müschen,Chih-Lin Hsieh,Michael R. Lieber +5 more
TL;DR: It is shown that although the CpG dinucleotide constitutes only 1% of the human genome, it accounts for 40%-70% of breakpoints at pro-B/pre-B stage translocation regions-specifically, those near the bcl-2, bCl-1, and E2A genes.
Journal ArticleDOI
XRCC4:DNA ligase IV can ligate incompatible DNA ends and can ligate across gaps
TL;DR: Human XRCC4:DNA ligase IV can ligate two double‐strand DNA ends that have fully incompatible short 3′ overhang configurations with no potential for base pairing, providing an explanation for several in vivo observations that were difficult to understand previously.
