Targeting mitochondrial biogenesis to overcome drug resistance to MAPK inhibitors
Gao Zhang,Dennie T. Frederick,Lawrence Wu,Zhi Wei,Clemens Krepler,Satish Srinivasan,Young Chan Chae,Xiaowei Xu,Harry Choi,Elaida Dimwamwa,Omotayo Ope,Batool Shannan,Devraj Basu,Devraj Basu,Dongmei Zhang,Manti Guha,Min Xiao,Sergio Randell,Katrin Sproesser,Wei Xu,Jephrey Y. Liu,Giorgos C. Karakousis,Lynn M. Schuchter,Tara C. Gangadhar,Ravi K. Amaravadi,Mengnan Gu,Caiyue Xu,Abheek Ghosh,Weiting Xu,Tian Tian,Jie Zhang,Shijie Zha,Qin Liu,Patricia Brafford,Ashani T. Weeraratna,Michael A. Davies,Jennifer A. Wargo,Narayan G. Avadhani,Yiling Lu,Gordon B. Mills,Dario C. Altieri,Keith T. Flaherty,Meenhard Herlyn +42 more
TL;DR: This study establishes mitochondrial biogenesis, coupled with aberrant tumor bioenergetics, as a potential therapy escape mechanism and paves the way for a rationale-based combinatorial strategy to improve the efficacy of MAPKi.
Journal ArticleDOI
Association of Biologic Therapy With Coronary Inflammation in Patients With Psoriasis as Assessed by Perivascular Fat Attenuation Index.
Youssef A. Elnabawi,Evangelos Oikonomou,Amit K. Dey,Jennifer Mancio,Justin A. Rodante,Milena Aksentijevich,Harry Choi,Andrew Keel,Julie Erb-Alvarez,Heather L. Teague,Aditya A. Joshi,Martin P. Playford,Benjamin Lockshin,Andrew D. Choi,Joel M. Gelfand,Marcus Y. Chen,David A. Bluemke,Cheerag Shirodaria,Charalambos Antoniades,Nehal N. Mehta +19 more
TL;DR: In this study, biologic therapy for moderate to severe psoriasis was associated with reduced coronary inflammation assessed by perivascular FAI, a novel imaging biomarker that assesses coronary inflammation by mapping spatial changes of periv vascular fat composition via coronary computed tomography angiography (CCTA).
Journal ArticleDOI
Tumor-associated B-cells induce tumor heterogeneity and therapy resistance
Rajasekharan Somasundaram,Gao Zhang,Mizuho Fukunaga-Kalabis,Michela Perego,Clemens Krepler,Xiaowei Xu,Christine Wagner,Denitsa Hristova,Jie Zhang,Tian Tian,Zhi Wei,Qin Liu,Kanika Garg,Johannes Griss,Rufus Hards,Margarita Maurer,Christine Hafner,Marius Mayerhöfer,Georgios Karanikas,Ahmad Jalili,Verena Bauer-Pohl,Felix Weihsengruber,Klemens Rappersberger,Josef Koller,Roland Lang,Courtney W. Schuchter,Dorothee Herlyn,Keith T. Flaherty,Meenhard Herlyn,Stephan N. Wagner +53 more
TL;DR: A mechanism of acquired drug resistance through the tumor microenvironment, which is mediated by human tumor-associated B cells-derived IGF-1 is described, with important clinical implications in melanoma patients.
Journal ArticleDOI
Treatment of Psoriasis With Biologic Therapy Is Associated With Improvement of Coronary Artery Plaque Lipid-Rich Necrotic Core: Results From a Prospective, Observational Study.
Harry Choi,Domingo E. Uceda,Amit K. Dey,Khaled Abdelrahman,Milena Aksentijevich,Justin A. Rodante,Youssef A. Elnabawi,Aarthi S Reddy,Andrew Keel,Julie Erb-Alvarez,Heather L. Teague,Martin P. Playford,Wunan Zhou,Marcus Y. Chen,Joel M. Gelfand,David A. Bluemke,Andrew J. Buckler,Nehal N. Mehta +17 more
TL;DR: Evidence of potential reduction in Lipid-rich necrotic core with treatment of systemic inflammation is provided and there was favorable modification of LRNC in those on biologic therapy.
Journal ArticleDOI
Association Between Soluble Lectinlike Oxidized Low-Density Lipoprotein Receptor-1 and Coronary Artery Disease in Psoriasis.
Amit K. Dey,Ranjitha Gaddipati,Youssef A. Elnabawi,Emily L. Ongstad,Aditya Goyal,Jonathan H. Chung,Heather L. Teague,Justin A. Rodante,Aparna A Sajja,Alexander V. Sorokin,Sundus S. Karathanasis,Joel M. Gelfand,Ruchi Gupta,Nehal N. Mehta +36 more
TL;DR: Soluble lectinlike oxidized low-density lipoprotein receptor-1 levels were elevated in patients with psoriasis and were associated with severity of skin disease, suggesting that inflammatory sLOX-1 induction may modulate lipid-rich NCB independent of hyperlipidemia status.
