The Hallmarks of Aging

In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes.

For example, a key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process versus those that measure flux through the autophagy pathway (i.e., the complete process including the amount and rate of cargo sequestered and degraded). In particular, a block in macroautophagy that results in autophagosome accumulation must be differentiated from stimuli that increase autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. It is worth emphasizing here that lysosomal digestion is a stage of autophagy and evaluating its competence is a crucial part of the evaluation of autophagic flux, or complete autophagy.

Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. Along these lines, because of the potential for pleiotropic effects due to blocking autophagy through genetic manipulation, it is imperative to target by gene knockout or RNA interference more than one autophagy-related protein. In addition, some individual Atg proteins, or groups of proteins, are involved in other cellular pathways implying that not all Atg proteins can be used as a specific marker for an autophagic process. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular assays, we hope to encourage technical innovation in the field.

Guidelines for the use and interpretation of assays for monitoring autophagy

Preclinical Development of a Bcma Targeting Antibody-Drug Conjugate with Novel Payload for Multiple Myeloma Therapy

In structured-light systems, the lens distortions of the camera and the projector reduce the measurement accuracy when calibrated as a standard stereo-vision system. The conventional compensation via distortion coefficients reduces the error, but still leaves a significant residual. Recently, we proposed a hybrid calibration procedure that leverages the standard calibration approach to improve measurement accuracy. This hybrid procedure consisted of building a pixel-wise phase-to-coordinate mapping based on adjusted 3D data obtained from the standard stereo-vision method. Here, we show experimentally that the measurement accuracy can be significantly improved, even using the linear pinhole model and linear mapping functions. We then move to consider the nonlinear model to improve the measurement accuracy further. Encouraging results show that this new calibration method increases the measurement accuracy without requiring elaborate calibration procedures or sophisticated ancillary equipment.

Toward high accuracy measurements in structured light systems

Zwitterionic polymers as crucial antifouling materials exhibit excellent antifouling performance due to their strong hydration ability. The structure–property relationship at the molecular level still remains to be elucidated. In this work, the surface hydration ability of three antifouling polymer membranes grafting on polysiloxane membranes Poly(sulfobetaine methacrylate) (T4-SB), poly(3-(methacryloyloxy)propane-1-sulfonate) (T4-SP), and poly(2-(dimethylamino)ethyl methacrylate) (T4-DM) was investigated. An orderly packed, and tightly bound surface hydration layer above T4-SP and T4-SB antifouling membranes was found by means of analyzing the dipole orientation distribution, diffusion coefficient, and average residence time. To further understand the surface hydration ability of three antifouling membranes, the surface structure, density profile, roughness, and area percentage of hydrophilic surface combining electrostatic potential, RDFs, SDFs, and noncovalent interactions of three polymers’ monomers were studied. It was concluded that the broadest distribution of electrostatic potential on the surface and the nature of anionic SO3- groups led to the following antifouling order of T4-SB > T4-SP > T4-DM. We hope that this work will gain some insight for the rational design and optimization of ecofriendly antifouling materials.

Molecular Dynamics Study on Properties of Hydration Layers above Polymer Antifouling Membranes

Radarsat-2 satellite offers general users the Quad-Pol SAR image service with a resolution of 8 meters, which provides valuable data source for the research of traffic vehicles monitoring on Quad-Pol SAR images. According to the vehicle target in such new SAR images, this paper put forward a target characteristic analysis method, which is a combination of target RCS measurement and polarization decomposition. Moreover, it choose the large trucks as an example, give out a conclusion of characteristic analysis of vehicle targets through the on-site synchronous experimental data obtained at different incident angle. Thus provides the research foundation for further exploration of realizing the traffic monitoring with space-born Quad-Pol SAR images.

Characteristic analysis of vehicle target in Quad-Pol Radarsat-2 SAR images

Ship wake detection can facilitate ship detection and ship motion parameters retrieval. In this paper, we propose a novel constant false alarm rate (CFAR) ship wake detection algorithm in SAR images based on length normalized scan method. As we all know, range component of ship's movement leads to azimuth offset in SAR images. The start point of wake should be located near the ship in the azimuth. According to this physical facts, length normalized scan is performed on line which may be a wake. By means of linear integral and length normalization technology, linear feature detection is transformed to point detection. Then the probability model in the scan domain is constructed to implement CFAR detection. Different resolution real SAR images are used to validate our proposed method. Experimental results show that this algorithm is effective for ship wake detection and can extract ship's velocity with good accuracy.
