Journal ArticleDOI
Mechanism of transmembrane signaling by sensor histidine kinases
Ivan Gushchin,Igor Melnikov,Vitaly Polovinkin,Vitaly Polovinkin,Andrii Ishchenko,Anastasia Yuzhakova,Pavel Buslaev,Gleb Bourenkov,Sergei Grudinin,Sergei Grudinin,Sergei Grudinin,Ekaterina Round,Taras Balandin,Valentin Borshchevskiy,Dieter Willbold,Gordon A. Leonard,Georg Büldt,Alexander Popov,Valentin Gordeliy,Valentin Gordeliy +19 more
TL;DR: The structures reveal the mechanism of transmembrane signal transduction in NarQ and show that binding of ligand induces displacement of the sensor domain helices by ~0.5 to 1 Å and demonstrate that the signaling-associated conformational changes in the TM domain do not need to be symmetric.
Journal ArticleDOI
Inward H$^+$ pump xenorhodopsin: Mechanism and alternative optogenetic approach
Vitaly Shevchenko,Vitaly Shevchenko,Thomas Mager,Kirill Kovalev,Kirill Kovalev,Vitaly Polovinkin,Vitaly Polovinkin,Alexey Alekseev,Alexey Alekseev,Josephine Juettner,Igor Chizhov,Christian Bamann,Charlotte D. Vavourakis,Rohit Ghai,Ivan Gushchin,Valentin Borshchevskiy,A. V. Rogachev,A. V. Rogachev,Igor Melnikov,Alexander Popov,Taras Balandin,Francisco Rodriguez-Valera,Dietmar J. Manstein,G. Bueldt,Ernst Bamberg,Valentin Gordeliy,Valentin Gordeliy +26 more
TL;DR: It is demonstrated that the NsXeR is a powerful pump, which is able to elicit action potentials in rat hippocampal neuronal cells up to their maximal intrinsic firing frequency, and they are an alternative to the well-known cation-selective channelrhodopsins.
Journal ArticleDOI
Fast iodide-SAD phasing for high-throughput membrane protein structure determination.
Igor Melnikov,Vitaly Polovinkin,Vitaly Polovinkin,Kirill Kovalev,Ivan Gushchin,Mikhail B. Shevtsov,Vitaly Shevchenko,Vitaly Shevchenko,Alexey Mishin,Alexey Alekseev,Francisco Rodriguez-Valera,Valentin Borshchevskiy,Vadim Cherezov,Vadim Cherezov,Gordon A. Leonard,Valentin Gordeliy,Valentin Gordeliy,Alexander Popov +17 more
TL;DR: A fast, easy, and potentially universal method for the de novo solution of the crystal structures of membrane proteins via iodide–single-wavelength anomalous diffraction (I-SAD), which can be applied using data collection strategies based on either standard or serial x-ray crystallography techniques.
Journal ArticleDOI
The complex analysis of X-ray mesh scans for macromolecular crystallography
TL;DR: A method and software program, MeshBest, for the detection of individual crystals based on two-dimensional X-ray mesh scans are presented.
Journal ArticleDOI
Sensor Histidine Kinase NarQ Activates via Helical Rotation, Diagonal Scissoring, and Eventually Piston-Like Shifts.
Ivan Gushchin,Philipp S. Orekhov,Igor Melnikov,Igor Melnikov,Vitaly Polovinkin,Vitaly Polovinkin,Vitaly Polovinkin,Anastasia Yuzhakova,Anastasia Yuzhakova,Valentin Gordeliy +9 more
TL;DR: It is concluded that helical rotation, diagonal scissoring, and piston are simply different degrees of freedom in coiled-coil proteins and are not mutually exclusive in NarQ, and likely in other nitrate sensors and TCS proteins as well.
