Human Homolog of patched, a Candidate Gene for the Basal Cell Nevus Syndrome
Ronald L. Johnson,Alana L. Rothman,Jingwu Xie,Lisa V. Goodrich,John W. Bare,Jeannette M. Bonifas,Anthony G. Quinn,Richard M. Myers,David R. Cox,Ervin H. Epstein,Matthew P. Scott +10 more
TL;DR: Heritable mutations in BCNS patients and a somatic mutation in a sporadic BCC were identified in a human homolog of the Drosophila patched (ptc) gene, which appears to be crucial for proper embryonic development and for tumor suppression.
Journal ArticleDOI
Activating Smoothened mutations in sporadic basal-cell carcinoma.
Jingwu Xie,Jingwu Xie,Maximilien Murone,Shiuh Ming Luoh,Anne M. Ryan,Qimin Gu,Chaohui Zhang,Jeannette M. Bonifas,Jeannette M. Bonifas,Ching-Wan Lam,Mary Hynes,Audrey Goddard,Arnon Rosenthal,Ervin H. Epstein,Ervin H. Epstein,Frederic J. de Sauvage +15 more
TL;DR: Findings support the role of SMO as a signalling component of the SHH–receptor complex and provide direct evidence that mutated SMO can function as an oncogene in BCCs.
Journal ArticleDOI
Activation of the hedgehog pathway in advanced prostate cancer
Tao Sheng,Chengxin Li,Xiaoli Zhang,Sumin Chi,Nonggao He,Kai Chen,Frank McCormick,Zoran Gatalica,Jingwu Xie +8 more
TL;DR: The data indicate that activation of the hedgehog pathway, through loss of Su(Fu) or overexpression of sonic hedgehog, may involve tumor progression and metastases of prostate cancer.
Journal Article
Mutations of the PATCHED Gene in Several Types of Sporadic Extracutaneous Tumors
Jingwu Xie,Ronald L. Johnson,Xiaoli Zhang,John W. Bare,Fred Waldman,Philip H. Cogen,Anil G. Menon,Robert S. Warren,Ling Chun Chen,Matthew P. Scott,Ervin H. Epstein +10 more
TL;DR: It is found that 2 of 14 sporadic medulloblastomas bear somatic nonsense mutations in one copy of the gene and also deletion of the other copy, and that missense mutations in PTCH are identified in two of seven breast carcinomas, one of nine meningiomas, and one colon cancer cell line.
Journal ArticleDOI
Activation of the hedgehog-signaling pathway in human cancer and the clinical implications
TL;DR: Major advances in the last 2 years in the understanding of hedgehog signaling activation in human cancer, interactions between Hedgehog signaling and other pathways in carcinogenesis, potential antagonists for hedgehog signaled inhibition and their clinical implications for human cancer treatment are summarized.
