SuFEx-enabled, agnostic discovery of covalent inhibitors of human neutrophil elastase

TL;DR: Evaluation of the ever-growing collection of SuFExable compounds toward various biological assays unexpectedly revealed a selective and covalent hNE inhibitor: benzene-1,2-disulfonyl fluoride.
Journal ArticleDOI

Sulfur(VI) Fluoride Exchange (SuFEx)-Enabled High-Throughput Medicinal Chemistry

TL;DR: A high-throughput hit-to-lead process based on the biocompatible SuFEx click chemistry that succeeds on picomole scale for direct screening and can accelerate the development of robust biological probes and drug candidates is demonstrated.
