Rapid assembly of SARS-CoV-2 genomes reveals attenuation of the Omicron BA.1 variant through NSP6

TL;DR: In this article , a plasmid-based genome assembly and rescue strategy (pGLUE) was proposed to construct complete infectious viruses or noninfectious subgenomic replicons in a single ligation reaction with >80% efficiency.

Recyclable CRISPR/Cas9 mediated gene disruption and deletions in Histoplasma

TL;DR: In this paper , the authors adapted a CRISPR/Cas9 system to facilitate targeted gene disruption in Histoplasma with high efficiency, and used this system to disrupt a number of target genes including RYP2 and SRE1 where loss-of-function mutations could be monitored visually by colony morphology or color.
