Journal ArticleDOI
Image-based analysis of lipid nanoparticle–mediated siRNA delivery, intracellular trafficking and endosomal escape
Jerome Gilleron,William Querbes,Anja Zeigerer,Anna Borodovsky,Giovanni Marsico,Undine Schubert,Kevin Manygoats,Sarah Seifert,Cordula Andree,Martin Stöter,Hila Epstein-Barash,Ligang Zhang,Victor Koteliansky,Kevin Fitzgerald,Eugenio Fava,Marc Bickle,Yannis Kalaidzidis,Akin Akinc,Martin Maier,Marino Zerial +19 more
TL;DR: It is estimated that escape of siRNAs from endosomes into the cytosol occurs at low efficiency (1–2%) and only during a limited window of time when the LNPs reside in a specific compartment sharing early and late endosomal characteristics.
Journal ArticleDOI
Multivalent N-Acetylgalactosamine-Conjugated siRNA Localizes in Hepatocytes and Elicits Robust RNAi-Mediated Gene Silencing
Jayaprakash K. Nair,Jennifer L. S. Willoughby,Amy Chan,Klaus Charisse,Md. Rowshon Alam,Qianfan Wang,Menno Hoekstra,Pachamuthu Kandasamy,Alexander V Kel'in,Stuart Milstein,Nate Taneja,Jonathan O'Shea,Sarfraz Shaikh,Ligang Zhang,Ronald J. van der Sluis,Michael E. Jung,Akin Akinc,Renta Hutabarat,Satya Kuchimanchi,Kevin Fitzgerald,Tracy Zimmermann,Theo J.C. Van Berkel,Martin Maier,Kallanthottathil G. Rajeev,Muthiah Manoharan +24 more
TL;DR: The optimally chemically modified siRNA-GalNAc conjugates are hepatotropic and long-acting and have the potential to treat a wide range of diseases involving liver-expressed genes.
Journal ArticleDOI
A rapid two-step method for isolation of functional primary mouse hepatocytes: cell characterization and asialoglycoprotein receptor based assay development.
Mariano Severgnini,Jennifer Sherman,Alfica Sehgal,Narayanannair K. Jayaprakash,Justin Aubin,Gang Wang,Ligang Zhang,Chang G. Peng,Kristina Yucius,James Butler,Kevin Fitzgerald +10 more
TL;DR: A modified two-step method for the rapid isolation and characterization of mouse primary hepatocytes that results in high yields of viable cells is described and an in vitro screening method for siRNA drug candidates was successfully developed utilizing freshly isolated hepatocytes with minimum culture time.
Journal ArticleDOI
Lipophilic siRNAs mediate efficient gene silencing in oligodendrocytes with direct CNS delivery
Qingmin Chen,David Butler,William Querbes,Rajendra K. Pandey,Pei Ge,Martin Maier,Ligang Zhang,Kallanthottathil G. Rajeev,Lubomir Nechev,Victor Kotelianski,Muthiah Manoharan,Dinah W.Y. Sah +11 more
TL;DR: Findings indicate that cholesterol-conjugated siRNAs are effective for direct CNS delivery to oligodendrocytes, and that the biocleavable disulfide linker appears to be beneficial for improving the potency of silencing of target mRNA in vivo.
Journal ArticleDOI
Modulation of thermal stability can enhance the potency of siRNA
Haripriya Addepalli,Meena,Chang G. Peng,Gang Wang,Yupeng Fan,Klaus Charisse,K. Narayanannair Jayaprakash,Kallanthottathil G. Rajeev,Rajendra K. Pandey,Gary Lavine,Ligang Zhang,Kerstin Jahn-Hofmann,Philipp Hadwiger,Muthiah Manoharan,Martin Maier +14 more
TL;DR: It is shown that the potency of siRNA can be improved by modulating the internal thermodynamic stability profile with chemical modifications, and placement of thermally destabilizing modifications, such as non-canonical bases like 2,4-difluorotoluene or single base pair mismatches in the central region of the sense strand, significantly improve the potency.
