Sensory Neurons Co-opt Classical Immune Signaling Pathways to Mediate Chronic Itch
Landon K. Oetjen,Madison R. Mack,Jing Feng,Timothy M. Whelan,Haixia Niu,Changxiong J. Guo,Sisi Chen,Anna M. Trier,Amy Z. Xu,Shivani V. Tripathi,Jialie Luo,Xiaofei Gao,Lihua Yang,Samantha L. Hamilton,Peter L. Wang,Jonathan R. Brestoff,Richard Brasington,Andras Schaffer,Frank Brombacher,Chyi-Song Hsieh,Robert W. Gereau,Mark J. Miller,Zhou-Feng Chen,Hongzhen Hu,Steve Davidson,Qin Liu,Brian S. Kim +26 more
TL;DR: An evolutionarily conserved paradigm is revealed in which the sensory nervous system employs classical immune signaling pathways to influence mammalian behavior, including chronic itch.
Journal ArticleDOI
Activation of Mast-Cell-Expressed Mas-Related G-Protein-Coupled Receptors Drives Non-histaminergic Itch
James Meixiong,Michael Anderson,Nathachit Limjunyawong,Mark F Sabbagh,Eric Hu,Madison R. Mack,Landon K. Oetjen,Fang Wang,Brian S. Kim,Xinzhong Dong +9 more
TL;DR: It is reported that mast cell activation via Mrgprb2 evoked non-histaminergic itch in mice independently of the IgE-Fc epsilon RI (FcεRI)-histamine axis, suggesting that this pathway may represent a therapeutic target for treating ACD and mast-cell-associated itch disorders in which antihistamines are ineffective.
Journal ArticleDOI
Intercellular Mitochondria Transfer to Macrophages Regulates White Adipose Tissue Homeostasis and Is Impaired in Obesity.
Jonathan R. Brestoff,Craig B. Wilen,John R. Moley,Yongjia Li,Wei Zou,Nicole P. Malvin,Marina N. Rowen,Brian Saunders,Hongming Ma,Madison R. Mack,Barry L. Hykes,Dale R. Balce,Anthony Orvedahl,Jesse W. Williams,Nidhi Rohatgi,Xiaoyan Wang,Michael R. McAllaster,Scott A. Handley,Brian S. Kim,John G. Doench,Bernd H. Zinselmeyer,Michael S. Diamond,Herbert W. Virgin,Herbert W. Virgin,Andrew E. Gelman,Steven L. Teitelbaum +25 more
TL;DR: It is found that macrophages acquire mitochondria from neighboring adipocytes in vivo and that this process defines a transcriptionally distinct macrophage subpopulation.
Journal ArticleDOI
The Itch-Scratch Cycle: A Neuroimmune Perspective.
Madison R. Mack,Brian S. Kim +1 more
TL;DR: This review highlights recent advances in the understanding of the neuroimmunology of chronic itch centered around three key points of entry into the itch-scratch cycle: the epithelial barrier, the immune system, and the peripheral nervous system.
Journal ArticleDOI
Sensory TRP channels contribute differentially to skin inflammation and persistent itch.
Jing Feng,Pu Yang,Madison R. Mack,Dariia Dryn,Dariia Dryn,Jialie Luo,Xuan Gong,Xuan Gong,Shenbin Liu,Landon K. Oetjen,Alexander Zholos,Zhinan Mei,Shijin Yin,Brian S. Kim,Hongzhen Hu +14 more
TL;DR: It is shown that persistent itch requires both TRPA1 and TRPV1, while TRPv1 has a protective role against skin inflammation in mice, which may provide new insights into the pathophysiology and treatment of chronic itch and inflammation in ACD patients.
