Journal ArticleDOI
Assessment of genotype imputation performance using 1000 Genomes in African American studies.
Dana B. Hancock,Joshua L. Levy,Nathan C. Gaddis,Laura J. Bierut,Nancy L. Saccone,Grier P. Page,Eric O. Johnson +6 more
TL;DR: Imputation quality for most programs was reduced by the addition of more distantly related reference populations, due entirely to the introduction of low frequency SNPs (MAF≤2%) that are monomorphic in the more closely related panels.
Journal ArticleDOI
Interplay of genetic risk factors and parent monitoring in risk for nicotine dependence
Li-Shiun Chen,Eric O. Johnson,Naomi Breslau,Dorothy K. Hatsukami,Nancy L. Saccone,Richard A. Grucza,Jen C. Wang,Anthony L. Hinrichs,Louis Fox,Alison Goate,John P. Rice,Laura J. Bierut +11 more
TL;DR: The genetic risk of nicotine dependent associated with rs16969968 was modified by level of parent monitoring, while the genetic risk associated withrs3743078 was not, suggesting that the increased risk due to some genes may be mitigated by environmental factors such as parent monitoring.
Journal ArticleDOI
Gamma-aminobutyric acid receptor genes and nicotine dependence: evidence for association from a case-control study
Arpana Agrawal,Michele L. Pergadia,Scott F. Saccone,Anthony L. Hinrichs,Christina N. Lessov-Schlaggar,Nancy L. Saccone,Rosalind J. Neuman,Naomi Breslau,Eric O. Johnson,Dorothy K. Hatsukami,Grant W. Montgomery,Andrew C. Heath,Nicholas G. Martin,Alison Goate,John P. Rice,Laura J. Bierut,Pamela A. F. Madden +16 more
TL;DR: This study did not find evidence for an association between GABBR2 gene and nicotine dependence, but given the potential role of compounds that enhance GABAergic neurotransmission in smoking cessation research, these findings have enormous potential for informing the wider field of addiction research.
Journal ArticleDOI
Associations and interactions between SNPs in the alcohol metabolizing genes and alcoholism phenotypes in European Americans
Richard Sherva,John P. Rice,Rosalind J. Neuman,Nanette Rochberg,Nancy L. Saccone,Laura J. Bierut +5 more
TL;DR: The large effect of rs1229984 on alcohol behavior is replicated, and although not common (MAF = 4%), this polymorphism may be highly relevant from a public health perspective in European Americans and may also affect alcohol behavior but requires replication.
Journal ArticleDOI
Genetic variation (CHRNA5), medication (combination nicotine replacement therapy vs. varenicline), and smoking cessation
Li-Shiun Chen,Timothy B. Baker,Douglas E. Jorenby,Megan E. Piper,Nancy L. Saccone,Eric O. Johnson,Naomi Breslau,Dorothy K. Hatsukami,Robert M. Carney,Laura J. Bierut +9 more
TL;DR: Results from two complementary smoking cessation trials demonstrate inconsistent genetic results in the placebo arms, highlighting the need to compare the most effective pharmacotherapies with the same placebo control to establish pharmacogenetic evidence to aid decisions on medication choice for patients trying to quit smoking.
