CRISPR/Cas9-engineered inducible gametocyte producer lines as a valuable tool for Plasmodium falciparum malaria transmission research.
Sylwia D. Boltryk,Sylwia D. Boltryk,Armin Passecker,Armin Passecker,Arne Alder,Arne Alder,Eilidh Carrington,Eilidh Carrington,Marga van de Vegte-Bolmer,Geert-Jan van Gemert,Alex van der Starre,Hans-Peter Beck,Hans-Peter Beck,Robert W. Sauerwein,Taco W. A. Kooij,Nicolas M. B. Brancucci,Nicolas M. B. Brancucci,Nicholas I Proellochs,Tim-Wolf Gilberger,Tim-Wolf Gilberger,Till S. Voss,Till S. Voss +21 more
TL;DR: In this paper, P. falciparum NF54 inducible gametocyte producer (iGP) lines for the routine mass production of synchronous gametocytes via conditional overexpression of the sexual commitment factor GDV1.
Journal ArticleDOI
Identification of essential exported Plasmodium falciparum protein kinases in malaria-infected red blood cells.
Ghizal Siddiqui,Ghizal Siddiqui,Nicholas I Proellochs,Nicholas I Proellochs,Brian M. Cooke,Brian M. Cooke +5 more
TL;DR: The diverse localisation and essentiality of these FIKKs demonstrate that they play different but essential roles in the survival of P. falciparum in RBCs and therefore are attractive new drug targets for the prevention or treatment of falcIParum malaria.
Journal ArticleDOI
A Prioritized and Validated Resource of Mitochondrial Proteins in Plasmodium Identifies Unique Biology.
Selma L van Esveld,Lisette Meerstein-Kessel,Cas Boshoven,Jochem F Baaij,Konstantin Barylyuk,Jordy P. M. Coolen,Joeri van Strien,Ronald A J Duim,Bas E. Dutilh,Daniel R. Garza,Marijn Letterie,Nicholas I Proellochs,Michelle N de Ridder,Prashanna Balaji Venkatasubramanian,Laura E. de Vries,Ross F. Waller,Taco W. A. Kooij,Martijn A. Huynen +17 more
TL;DR: In this article, a comprehensive list of Plasmodium falciparum mitochondrial proteins is presented, including proteins that have not been identified as mitochondrial in other eukaryotes but have characterized homologs in bacteria that are involved in metabolism or translation.
Journal ArticleDOI
The PHIST protein GEXP02 targets the host cytoskeleton during sexual development of Plasmodium falciparum.
Jan D. Warncke,Jan D. Warncke,Armin Passecker,Armin Passecker,Enja Kipfer,Enja Kipfer,Enja Kipfer,Françoise Brand,Françoise Brand,Lara Pérez-Martínez,Nicholas I Proellochs,Taco W. A. Kooij,Falk Butter,Till S. Voss,Till S. Voss,Hans-Peter Beck,Hans-Peter Beck +16 more
TL;DR: It is shown by immunofluorescence that GEXP02 is exported to the gametocyte‐infected host cell periphery, which indicates that G EXP02 targets the erythrocyte cytoskeleton and is likely involved in its remodelling.
Journal ArticleDOI
Novel insights from the Plasmodium falciparum sporozoite-specific proteome by probabilistic integration of 26 studies.
Lisette Meerstein-Kessel,Jeron Venhuizen,Daniel R. Garza,Nicholas I Proellochs,Emma J. Vos,Joshua M. Obiero,Philip L. Felgner,Robert W. Sauerwein,Marynthe Peters,Annie S. P. Yang,Martijn A. Huynen +10 more
TL;DR: In this paper, the authors applied Bayesian data integration to determine which proteins are not only present in sporozoites but are also specific to that stage, and found that sporozoite proteins are enriched for proteins involved in type II fatty acid synthesis in the apicoplast and GPI anchor synthesis.
