Journal ArticleDOI
Staphylococcus aureus resistance to human defensins and evasion of neutrophil killing via the novel virulence factor MprF is based on modification of membrane lipids with L-lysine
Andreas Peschel,Andreas Peschel,Ralph W. Jack,Michael Otto,L. Vincent Collins,Petra Staubitz,Graeme J. Nicholson,Hubert Kalbacher,Willem F. Nieuwenhuizen,Günther Jung,Andrej Tarkowski,Kok P. M. van Kessel,Jos A. G. van Strijp +12 more
TL;DR: A novel staphylococcal gene, mprF, which determines resistance to several host defense peptides such as defensins and protegrins constitutes a novel virulence factor, which may be of general relevance for bacterial pathogens and represents a new target for attacking multidrug resistant bacteria.
Journal ArticleDOI
The bacterial defensin resistance protein MprF consists of separable domains for lipid lysinylation and antimicrobial peptide repulsion.
Christoph M. Ernst,Petra Staubitz,Nagendra N. Mishra,Soo-Jin Yang,Gabriele Hornig,Hubert Kalbacher,Arnold S. Bayer,Dirk Kraus,Andreas Peschel +8 more
TL;DR: It is demonstrated that the hydrophilic C-terminal domain and six of the fourteen proposed trans-membrane segments of MprF are sufficient for full-level lysyl-phosphatidylglycerol (Lys-PG) production and that several conserved amino acid positions in MPRF are indispensable for Lys-PG production.
Journal ArticleDOI
MprF-mediated biosynthesis of lysylphosphatidylglycerol, an important determinant in staphylococcal defensin resistance.
TL;DR: It is demonstrated here that expression of mprF is sufficient to confer L-PG production in Escherichia coli, which indicates that MprF represents the L-pg synthase, and forms the basis for further detailed analyses of L- PG biosynthesis and its role in bacterial infections.
Journal ArticleDOI
Structure-function relationships in the tryptophan-rich, antimicrobial peptide indolicidin.
Petra Staubitz,Andreas Peschel,Willem F. Nieuwenhuizen,Michael Otto,Friedrich Götz,Günther Jung,Ralph W. Jack +6 more
TL;DR: Indolicidin is a cationic 13 amino acid peptide amide produced in the granules of bovine neutrophils with the sequence H-ILPWKWPWWPWRR-NH2 as discussed by the authors.
Journal ArticleDOI
In vitro susceptibility of Staphylococcus aureus to thrombin-induced platelet microbicidal protein-1 (tPMP-1) is influenced by cell membrane phospholipid composition and asymmetry.
Kasturi Mukhopadhyay,William A. Whitmire,Yan Q. Xiong,Yan Q. Xiong,Jaime Molden,Tiffanny Jones,Andreas Peschel,Petra Staubitz,Jill Adler-Moore,Peter J. McNamara,Richard A. Proctor,Michael R. Yeaman,Michael R. Yeaman,Arnold S. Bayer,Arnold S. Bayer +14 more
TL;DR: In this paper, the content and distribution of phospholipid (PL) constituents were compared in an isogenic tPMP-susceptible (tPMP(S)) and -resistant (tMPP(R)) S. aureus strain pair (ISP479C vs ISP479R respectively).
