Amyloid β Protein and Alzheimer’s Disease: When Computer Simulations Complement Experimental Studies
Jessica Nasica-Labouze,Phuong H. Nguyen,Fabio Sterpone,Olivia Berthoumieu,Nicolae-Viorel Buchete,Sébastien Côté,Alfonso De Simone,Andrew J. Doig,Peter Faller,Angel E. Garcia,Alessandro Laio,Mai Suan Li,Simone Melchionna,Normand Mousseau,Yuguang Mu,Anant K. Paravastu,Samuela Pasquali,David J. Rosenman,Birgit Strodel,Bogdan Tarus,John H. Viles,Tong Zhang,Tong Zhang,Chunyu Wang,Philippe Derreumaux,Philippe Derreumaux +25 more
TL;DR: Simulations Complement Experimental Studies Jessica Nasica-Labouze, Phuong H. Nguyen, Fabio Sterpone,† Olivia Berthoumieu,‡ Nicolae-Viorel Buchete, Sebastien Cote, Alfonso De Simone, Andrew J. Doig, and Philippe Derreumaux are authors of this paper.
Journal ArticleDOI
Dihedral angle principal component analysis of molecular dynamics simulations
TL;DR: It is shown that the dPCA amounts to a one-to-one representation of the original angle distribution and that its principal components can readily be characterized by the corresponding conformational changes of the peptide.
Journal ArticleDOI
Amyloid oligomers: A joint experimental/computational perspective on Alzheimer's disease, Parkinson's disease, type II diabetes, and amyotrophic lateral sclerosis
Phuong H. Nguyen,Ayyalusamy Ramamoorthy,Bikash R. Sahoo,Jie Zheng,Peter Faller,John E. Straub,Laura Dominguez,Joan-Emma Shea,Nikolay V. Dokholyan,Alfonso De Simone,Alfonso De Simone,Buyong Ma,Buyong Ma,Ruth Nussinov,Saeed Najafi,Son Tung Ngo,Antoine Loquet,Mara Chiricotto,Pritam Ganguly,James McCarty,Mai Suan Li,Carol K. Hall,Yiming Wang,Yifat Miller,Simone Melchionna,Birgit Habenstein,Stepan Timr,Jiaxing Chen,Brianna Hnath,Birgit Strodel,Rakez Kayed,Sylvain Lesné,Guanghong Wei,Fabio Sterpone,Andrew J. Doig,Philippe Derreumaux,Philippe Derreumaux +36 more
TL;DR: In this paper, the authors review what computer, in vitro, in vivo, and pharmacological experiments tell us about the accumulation and deposition of the oligomers of the (Aβ, tau), α-synuclein, IAPP, and superoxide dismutase 1 proteins, which have been the mainstream concept underlying Alzheimer's disease, Parkinson's disease (PD), type II diabetes (T2D), and amyotrophic lateral sclerosis (ALS) research.
Journal ArticleDOI
Energy transport in peptide helices
Virgiliu Botan,Ellen H. G. Backus,Rolf Pfister,Alessandro Moretto,Marco Crisma,Claudio Toniolo,Phuong H. Nguyen,Gerhard Stock,Peter Hamm +8 more
TL;DR: Energy transport through an α-aminoisobutyric acid-based 310-helix dissolved in chloroform is investigated in a combined experimental-theoretical approach, revealing good qualitative agreement with experiment but in terms of quantitative numbers, theory and experiment differ.
Journal ArticleDOI
Why Is Research on Amyloid-β Failing to Give New Drugs for Alzheimer’s Disease?
Andrew J. Doig,Maria P. del Castillo-Frias,Olivia Berthoumieu,Olivia Berthoumieu,Bogdan Tarus,Jessica Nasica-Labouze,Fabio Sterpone,Phuong H. Nguyen,Nigel M. Hooper,Peter Faller,Peter Faller,Philippe Derreumaux +11 more
TL;DR: From a review of the recent literature and the own experience based on in vitro, in silico, and in vivo studies, some reasons to explain this repetitive failure of drugs targeting Aβ are presented.
