Spectinamides: a new class of semisynthetic antituberculosis agents that overcome native drug efflux
Richard E. Lee,Julian G. Hurdle,Jiuyu Liu,David F. Bruhn,Tanja Matt,Michael S. Scherman,Pavan K. Vaddady,Zhong Zheng,Jianjun Qi,Rashid Akbergenov,Sourav Das,Dora B. Madhura,Chetan Rathi,Ashit Trivedi,Cristina Villellas,Robin B. Lee,Rakesh,Samanthi L. Waidyarachchi,Dianqing Sun,Michael R. McNeil,José A. Aínsa,Helena I. Boshoff,Mercedes Gonzalez-Juarrero,Bernd Meibohm,Erik C. Böttger,Anne J. Lenaerts +25 more
TL;DR: The antitubercular efficacy of spectinamides demonstrates that synthetic modifications to classical antibiotics can overcome the challenge of intrinsic efflux pump-mediated resistance and expands opportunities for target-based tuberculosis drug discovery.
Journal ArticleDOI
Structure-Activity Relationships of Spectinamide Antituberculosis Agents: A Dissection of Ribosomal Inhibition and Native Efflux Avoidance Contributions.
Jiuyu Liu,David F. Bruhn,Robin B. Lee,Zhong Zheng,Tanja Janusic,Dimitri Scherbakov,Michael S. Scherman,Helena I. Boshoff,Sourav Das,Rakesh,Samanthi L. Waidyarachchi,Tiffany A. Brewer,Tiffany A. Brewer,Begoña Gracia,Lei Yang,John C. Bollinger,Gregory T. Robertson,Bernd Meibohm,Anne J. Lenaerts,José A. Aínsa,Erik C. Böttger,Richard E. Lee +21 more
TL;DR: The structure-activity relationships detailed here emphasize the need to examine efflux-mediated resistance in the design of antituberculosis drugs and demonstrate that it is possible to overcome intrinsic efflux with synthetic modification.
Journal ArticleDOI
Aminomethyl Spectinomycins as Therapeutics for Drug-Resistant Gonorrhea and Chlamydia Coinfections.
Michelle M. Butler,Samanthi L. Waidyarachchi,Kristie L. Connolly,Ann E. Jerse,Weirui Chai,Richard E. Lee,Stephan Kohlhoff,Dean L. Shinabarger,Terry L. Bowlin +8 more
TL;DR: Data produced by this study support aminomethyl spectinomycins as a promising replacement for spect inomycin and antibiotics such as ceftriaxone for treating drug-resistant gonorrhea, with the added benefit of treating chlamydial coinfections.
Journal ArticleDOI
Aminomethyl spectinomycins as therapeutics for drug-resistant respiratory tract and sexually transmitted bacterial infections
David F. Bruhn,Samanthi L. Waidyarachchi,Dora B. Madhura,Dimitri Shcherbakov,Zhong Zheng,Jiuyu Liu,Yasser M. AbdelRahman,Aman P. Singh,Aman P. Singh,Stefan Duscha,Chetan Rathi,Robin B. Lee,Robert J. Belland,Bernd Meibohm,Jason W. Rosch,Erik C. Böttger,Richard E. Lee +16 more
TL;DR: A new series of spectinomycin analogs with potency against drug-resistant bacterial pathogens was designed and developed with a structure-based approach and validated in vitro and in vivo, indicating that N-benzyl aminomethyl spect inomycins are suitable for further development to treat drug- resistant respiratory tract and sexually transmitted bacterial infections.
Journal ArticleDOI
Aminomethyl spectinomycins: a novel antibacterial chemotype for biothreat pathogens.
Jennifer M. Scarff,Samanthi L. Waidyarachchi,Christopher J. Meyer,Douglas Lane,Weirui Chai,Margaret M. Lemmon,Jiuyu Liu,Michelle M. Butler,Terry L. Bowlin,Richard E. Lee,Rekha G. Panchal +10 more
TL;DR: Aminomethyl spectinomycins are promising new candidates for development of therapeutics against biodefense bacterial agents against multi-drug-resistant strains and difficult-to-treat bacterial infections.
