Journal ArticleDOI
Genomewide association analysis of coronary artery disease.
Nilesh J. Samani,Jeanette Erdmann,Alistair S. Hall,Christian Hengstenberg,Massimo Mangino,Bjoern Mayer,Richard J. Dixon,Thomas Meitinger,Peter S. Braund,Jennifer H. Barrett,Inke R. König,Suzanne Stevens,Silke Szymczak,David-Alexandre Trégouët,Mark M. Iles,Friedrich Pahlke,Helen Pollard,Wolfgang Lieb,François Cambien,Marcus Fischer,Willem H. Ouwehand,Stefan Blankenberg,Anthony J. Balmforth,Andrea Baessler,Stephen G. Ball,Tim M. Strom,Ingrid Braenne,Christian Gieger,Panos Deloukas,Martin D. Tobin,Andreas Ziegler,John R. Thompson,Heribert Schunkert +32 more
TL;DR: Several genetic loci that, individually and in aggregate, substantially affect the risk of development of coronary artery disease are identified.
Journal ArticleDOI
Genetics and Beyond – The Transcriptome of Human Monocytes and Disease Susceptibility
Tanja Zeller,Philipp S. Wild,Silke Szymczak,Maxime Rotival,Arne Schillert,Raphaële Castagné,Seraya Maouche,Marine Germain,Karl J. Lackner,Heidi Rossmann,Medea S. Eleftheriadis,C. Sinning,Renate B. TL;DR: This study demonstrates that the monocyte transcriptome is a potent integrator of genetic and non-genetic influences of relevance for disease pathophysiology and risk assessment.
Journal ArticleDOI
Genome-wide association analysis identifies variation in vitamin D receptor and other host factors influencing the gut microbiota
Jun Wang,Louise B. Thingholm,Jurgita Skiecevičienė,Philipp Rausch,Philipp Rausch,Martin Kummen,Johannes R. Hov,Frauke Degenhardt,Femke-Anouska Heinsen,Malte C. Karlsen,John F. Baines,John F. Baines,Andre Franke +42 more
TL;DR: Genome-wide significant associations for overall microbial variation and individual taxa at multiple genetic loci identify other important points of host–microbe intersection, notably several disease susceptibility genes and sterol metabolism pathway components.
Journal ArticleDOI
Evaluation of variable selection methods for random forests and omics data sets.
TL;DR: Vita is considerably faster than Boruta and thus more suitable for large data sets, but only Boruta can also be applied in low-dimensional settings, while Vita was the most robust approach under a pure null model without any predictor variables related to the outcome.
Journal ArticleDOI
A trans-acting locus regulates an anti-viral expression network and type 1 diabetes risk
Matthias Heinig,Enrico Petretto,Chris Wallace,Leonardo Bottolo,Maxime Rotival,Han Lu,Yoyo Li,Rizwan Sarwar,Sarah R. Langley,Anja Bauerfeind,Oliver Hummel,Young-Ae Lee,Young-Ae Lee,Svetlana Paskas,Carola Rintisch,Kathrin Saar,Jason D. Cooper,Rachel Buchan,Elizabeth E. Gray,Jason G. Aitman,François Cambien,David Clayton,John A. Todd,Norbert Hubner,Norbert Hubner,Stuart A. Cook,Stuart A. Cook +47 more
TL;DR: The use of integrated genome-wide approaches across seven rat tissues to identify gene networks and the loci underlying their regulation implicate IRF7 network genes and their regulatory locus in the pathogenesis of T1D.
