Cholesterol uptake disruption, in association with chemotherapy, is a promising combined metabolic therapy for pancreatic adenocarcinoma
Fabienne Guillaumond,Ghislain Bidaut,Mehdi Ouaissi,Stéphane Servais,Victoire Gouirand,Orianne Olivares,Sophie Lac,Laurence Borge,Julie Roques,Odile Gayet,Michelle Pinault,Cyrille Guimaraes,Jérémy Nigri,Celine Loncle,Marie-Noëlle Lavaut,Stéphane Garcia,Anne Tailleux,Bart Staels,Ezequiel Calvo,Richard Tomasini,Juan L. Iovanna,Sophie Vasseur +21 more
TL;DR: E elucidation of the metabolic signature of PDAC has identified the low-density lipoprotein receptor (LDLR), which facilitates cholesterol uptake, as a promising therapeutic target, and identifies LDLR as a novel metabolic target to limit PDAC progression and disease patient relapse.
Journal ArticleDOI
Collagen-derived proline promotes pancreatic ductal adenocarcinoma cell survival under nutrient limited conditions.
Orianne Olivares,Jared R. Mayers,Victoire Gouirand,Margaret E. Torrence,Tristan Gicquel,Laurence Borge,Sophie Lac,Julie Roques,Marie-Noëlle Lavaut,Patrice Berthezene,Marion Rubis,Véronique Secq,Stéphane Garcia,Vincent Moutardier,Dominique Lombardo,Juan L. Iovanna,Richard Tomasini,Fabienne Guillaumond,Matthew G. Vander Heiden,Matthew G. Vander Heiden,Sophie Vasseur +20 more
TL;DR: It is demonstrated that collagen serves as a proline reservoir for PDAC cells to use as a nutrient source when other fuels are limited and that proline oxidase (PRODH1) is required forPDAC cell proliferation in vitro and in vivo.
Journal ArticleDOI
Cancer-associated fibroblast-derived annexin A6+ extracellular vesicles support pancreatic cancer aggressiveness
Julie Leca,Sébastien Martinez,Sophie Lac,Jérémy Nigri,Véronique Secq,Marion Rubis,Christian Bressy,Arnauld Sergé,Marie-Noëlle Lavaut,Nelson Dusetti,Celine Loncle,Julie Roques,Daniel Pietrasz,Corinne Bousquet,Stéphane Garcia,Samuel Granjeaud,Mehdi Ouaissi,Jean-Baptiste Bachet,Christine Brun,Juan L. Iovanna,Pascale Zimmermann,Sophie Vasseur,Richard Tomasini +22 more
TL;DR: It is suggested that CAF-tumor cell crosstalk supported by ANXA6+ EVs is predictive of PDA aggressiveness, highlighting a therapeutic target and potential biomarker for PDA.
Journal ArticleDOI
TAp73 is required for spermatogenesis and the maintenance of male fertility.
Satoshi Inoue,Richard Tomasini,Richard Tomasini,Alessandro Rufini,Andrew J. Elia,Massimiliano Agostini,Ivano Amelio,Dave Cescon,David Dinsdale,Lily Zhou,Isaac S. Harris,Sophie Lac,Jennifer Silvester,Wanda Y. Li,Masato Sasaki,Jillian Haight,Anne Brüstle,Andrew Wakeham,Colin McKerlie,Andrea Jurisicova,Gerry Melino,Gerry Melino,Tak W. Mak +22 more
TL;DR: It is demonstrated that deficiency of the TAp73 isoform, but not p53 or ΔNp73, leads to male infertility caused by severely impaired germ cell differentiation and maturation to viable sperms in the testes and previously unidentified in vivo evidence that TAp 73 has a unique role in spermatogenesis that ensures the maintenance of mitotic cells and normal spermiogenesis.
Journal ArticleDOI
IL17 Functions through the Novel REG3β–JAK2–STAT3 Inflammatory Pathway to Promote the Transition from Chronic Pancreatitis to Pancreatic Cancer
Celine Loncle,Laia Bonjoch,Emma Folch-Puy,Maria Belen Lopez-Millan,Sophie Lac,Maria Ines Molejon,Eduardo Chuluyan,Pierre Cordelier,Pierre Dubus,Gwen Lomberk,Raul Urrutia,Daniel Closa,Juan L. Iovanna +12 more
TL;DR: It is demonstrated that IL17 induces the expression of REG3β, a well-known mediator of pancreatitis, during acinar-to-ductal metaplasia and in early pancreatic intraepithelial neoplasia (PanIN) lesions and that REG3 β promotes cell growth and decreases sensitivity to cell death through activation of the gp130-JAK2-STAT3-dependent pathway.
