Journal ArticleDOI
Lipid metabolic reprogramming in cancer cells
TL;DR: In it, detailed insight is provided into the essential roles exerted by specific lipids in mediating intracellular oncogenic signaling, endoplasmic reticulum stress and bidirectional crosstalk between cells of the tumor microenvironment and cancer cells.
Journal ArticleDOI
Strengthened glycolysis under hypoxia supports tumor symbiosis and hexosamine biosynthesis in pancreatic adenocarcinoma
Fabienne Guillaumond,Julie Leca,Orianne Olivares,Marie Noelle Lavaut,Nicolas Vidal,Patrice Berthezene,Nelson Dusetti,Celine Loncle,Ezequiel Calvo,Olivier Turrini,Juan L. Iovanna,Richard Tomasini,Sophie Vasseur +12 more
TL;DR: It is reported that hypoxia increases transcription of hexosamine biosynthetic pathway genes as well as levels of O-glycosylated proteins and that O-linked N-acetylglucosaminylation of proteins is a process required for hypoxic pancreatic cancer cell survival.
Journal ArticleDOI
Cholesterol uptake disruption, in association with chemotherapy, is a promising combined metabolic therapy for pancreatic adenocarcinoma
Fabienne Guillaumond,Ghislain Bidaut,Mehdi Ouaissi,Stéphane Servais,Victoire Gouirand,Orianne Olivares,Sophie Lac,Laurence Borge,Julie Roques,Odile Gayet,Michelle Pinault,Cyrille Guimaraes,Jérémy Nigri,Celine Loncle,Marie-Noëlle Lavaut,Stéphane Garcia,Anne Tailleux,Bart Staels,Ezequiel Calvo,Richard Tomasini,Juan L. Iovanna,Sophie Vasseur +21 more
TL;DR: E elucidation of the metabolic signature of PDAC has identified the low-density lipoprotein receptor (LDLR), which facilitates cholesterol uptake, as a promising therapeutic target, and identifies LDLR as a novel metabolic target to limit PDAC progression and disease patient relapse.
Journal ArticleDOI
Collagen-derived proline promotes pancreatic ductal adenocarcinoma cell survival under nutrient limited conditions.
Orianne Olivares,Jared R. Mayers,Victoire Gouirand,Margaret E. Torrence,Tristan Gicquel,Laurence Borge,Sophie Lac,Julie Roques,Marie-Noëlle Lavaut,Patrice Berthezene,Marion Rubis,Véronique Secq,Stéphane Garcia,Vincent Moutardier,Dominique Lombardo,Juan L. Iovanna,Richard Tomasini,Fabienne Guillaumond,Matthew G. Vander Heiden,Matthew G. Vander Heiden,Sophie Vasseur +20 more
TL;DR: It is demonstrated that collagen serves as a proline reservoir for PDAC cells to use as a nutrient source when other fuels are limited and that proline oxidase (PRODH1) is required forPDAC cell proliferation in vitro and in vivo.
Journal ArticleDOI
Cloning and Expression of the Rat p8 cDNA, a New Gene Activated in Pancreas during the Acute Phase of Pancreatitis, Pancreatic Development, and Regeneration, and Which Promotes Cellular Growth
Gustavo V. Mallo,Fritz Fiedler,Ezequiel Calvo,Emilia M. Ortiz,Sophie Vasseur,Volker Keim,Jean Morisset,Juan L. Iovanna +7 more
TL;DR: Analysis of p8's primary and secondary structure suggests that p8 is a basic helix-turn-helix-containing gene with slight homology to several homeotic genes and sufficient signal to be targeted to the nucleus, and it is proposed p8 as a putative transcriptional factor which can regulate pancreatic growth.
