DNA Methylation Profiling of Uniparental Disomy Subjects Provides a Map of Parental Epigenetic Bias in the Human Genome
Ricky S. Joshi,Paras Garg,Noah Zaitlen,Tuuli Lappalainen,Corey T. Watson,Nidha Azam,Daniel Ho,Xin Li,Stylianos E. Antonarakis,Han G. Brunner,Karin Buiting,Sau Wai Cheung,Bradford Coffee,Thomas Eggermann,David Francis,Joep P.M. Geraedts,Giorgio Gimelli,Samuel G. Jacobson,Cédric Le Caignec,Cédric Le Caignec,Nicole de Leeuw,Thomas Liehr,Deborah J G Mackay,Stephen B. Montgomery,Alistair T. Pagnamenta,Peter Papenhausen,David O. Robinson,Claudia A. L. Ruivenkamp,Charles E. Schwartz,Bernhard Steiner,David A. Stevenson,Urvashi Surti,Thomas H. Wassink,Andrew J. Sharp +33 more
TL;DR: The data provide a detailed map of parental epigenetic bias in the human genome, providing insights into potential parent-of-origin effects and patterns of epigenetic variation at DMRs.
Journal ArticleDOI
Brief Report: Isogenic Induced Pluripotent Stem Cell Lines From an Adult With Mosaic Down Syndrome Model Accelerated Neuronal Ageing and Neurodegeneration
Aoife Murray,Audrey Letourneau,Claudia Canzonetta,Elisavet Stathaki,Stefania Gimelli,Frédérique Sloan-Béna,Robert Abrehart,Pollyanna Goh,Shuhui Lim,C. Baldo,Franca Dagna-Bricarelli,Saad Hannan,Martin Mortensen,David Ballard,Denise Syndercombe Court,Noemi Fusaki,Mamoru Hasegawa,Trevor G. Smart,Cleo L. Bishop,Stylianos E. Antonarakis,Jürgen Groet,Jürgen Groet,Dean Nizetic,Dean Nizetic +23 more
TL;DR: This work has developed a first nonintegration‐reprogrammed isogenic human induced pluripotent stem cell (iPSC) model of DS by reprogramming the skin fibroblasts from an adult individual with constitutional mosaicism for DS and separately cloning multiple isogenic T21 and euploid (D21) iPSC lines.
Journal ArticleDOI
Cardiomyogenesis is controlled by the miR-99a/let-7c cluster and epigenetic modifications.
Antonietta Coppola,Antonio Romito,Christelle Borel,Corinne Gehrig,Maryline Gagnebin,Emilie Falconnet,Antonella Izzo,Lucia Altucci,Sandro Banfi,Stylianos E. Antonarakis,Gabriella Minchiotti,Gilda Cobellis +11 more
TL;DR: It is shown that the miR-99a/let-7c cluster, mapping on human chromosome 21, is involved in the control of cardiomyogenesis by altering epigenetic factors, and it is demonstrated that the identified targets are underexpressed in human Down syndrome fetal heart specimens.
Journal ArticleDOI
Network-Guided Analysis of Genes with Altered Somatic Copy Number and Gene Expression Reveals Pathways Commonly Perturbed in Metastatic Melanoma
Armand Valsesia,Armand Valsesia,Armand Valsesia,Donata Rimoldi,Danielle Martinet,Mark Ibberson,Paola Benaglio,Manfredo Quadroni,Patrice Waridel,Muriel Gaillard,Mireille Pidoux,Blandine Rapin,Carlo Rivolta,Ioannis Xenarios,Andrew J. Victor Jongeneel,Christian Iseli,Christian Iseli,Brian Stevenson,Brian Stevenson +24 more
TL;DR: The results emphasize the potential of the EPHA3 and FRS2 gene products, involved in angiogenesis and migration, as possible therapeutic targets in melanoma and demonstrates the utility of network-guided approaches, for both large and small datasets, to identify pathways recurrently perturbed in cancer.
