TL;DR: The identification of Smad3/Smad4 binding sequences, termed CAGA boxes, within the promoter of the human PAI‐1 gene is reported, which suggest that this may be a widely used motif in TGFβ‐regulated transcription.

TL;DR: In vitro studies indicate that a domain corresponding to exon 3 is responsible for the absence of Smad2 transcriptional activity in CAGA box-containing promoters, suggesting that Smad 2 and Smad3 may have different subsets of target genes participating thus in distinct responses among TGFβ pleiotropic effects.
