Abstract: Normal Japanese subjects were divided into two groups, ie, one with both low and high Km isozymes of aldehyde dehydrogenase for acetaldehyde, and the other deficient in the low Km isozyme After intake of 04 g/kg alcohol, the deficient subjects showed high level of blood acetaldehyde, facial flushing and the other dysphoric symptoms, including increase of pulse rate, decrease of diastolic blood pressure, changes of pulse wave in the fingertip, and elevation of the arterial pressure and blood flow rate in common carotid arteries as well as increase of plasma catecholamines level In contrast, subjects with normal ALDH did not show these changes From the observation of liver specimens obtained at autopsy, the frequency of deficient phenotype of ALDH in Japanese was presumed to be about 36%

Abstract: The presence of fibronectin (FN) and glial fibrillary acidic protein (GFAP) in two astrocytomas, 17 glioblastomas, and five meningiomas was studied by indirect immunoproxidase staining of formalin-fixed and paraffin-embedded surgical specimens. Angiogenesis in tumor was scored by the microscopic angiogenesis grading system (MAGS), and plasma FN levels were measured by single radial immunodiffusion. In astrocytomas and glioblastomas, GFAP-positive tumor cells had no FN expression and FN was confined to proliferating vessel walls and the leptomeninges, showing a mutually exclusive FN and GFAP expression. GFAP-positive tumor cells were occasionally surrounded by a network of FN-positive matrix produced by cells derived from the leptomeninges or blood vessels. In meningiomas, FN expression was found in vessel walls and meningioma cells including whorl formations and psammoma bodies. In general, deep immunoperoxidase staining for FN was shown in the endothelial cells and the psammoma bodies. Plasma FN levels were correlated significantly not to the degree of leptomeningeal proliferation but to the MAGS scores in gliomas.

Abstract: We have examined mast-cell precursors in various in vivo and in vitro haematopoietic colonies of mice. Cells from haematopoietic colonies of (WB x C57BL/6)F1- + +/+ mouse origin were injected into the skin of the congeneic W/Wv mice which are genetically depleted of tissue mast cells. The appearance of mast-cell clusters at the injection site indicated the presence of mast-cell precursors in the injected cell suspension. More than 40% of 12 d exogenous spleen colonies and 14 d in vitro mixed colonies contained mast-cell precursors, but only a small proportion of 7 d exogenous spleen colonies, 9 d in vitro erythroid bursts or 14 d large in vitro neutrophil-macrophage colonies contained mast-cell precursors. Neither transient endogenous erythroid spleen colonies examined at the fifth day nor 7 d in vitro neutrophil-macrophage colonies contained mast-cell precursors. Among 12 d spleen colonies mast-cell precursors were more frequent in the predominantly neutrophil than the predominantly erythroid colonies but 14 d in vitro mixed colonies, in which erythroid cells were predominant, contained mast-cell precursors more frequently than 14 d in vitro neutrophil-macrophage colonies. Therefore, differentiation of mast cells seems to be independent of either the neutrophil-macrophage or erythroid cell lineages.

Abstract: The structure and location of the dorsal and ventral tegmental nuclei of Gudden were studied in brains of 12 species of mammals. The ventral teg-mental nucleus of Gudden is composed of oval or polygonal, medium-sized, well-stained cells, and includes the pars principalis situated ventral to the fasciculus longitudinalis medialis, and the pars suprafascicularis in the cen-tral gray matter dorsal to the fasciculus longitudinalis medialis (in the golden hamster, mouse, vole, and house shrew). The dorsal tegmental nucleus of Gudden is situated within the central gray matter. This nucleus includes the pars ventralis, composed of oval or triangular cells, medium-sized but smaller than those of the ventral tegmen-tal nucleus of Gudden, and the pars dorsalis, composed of round, small, and lightly stained cells, and often encapsulated by fibers of the fasciculus longi-tudinalis dorsalis. The isthmus between the dorsal and ventral tegmental nuclei of Gudden is distinct. In the cat, dog, rat, gerbil, and rabbit, the isthmus stands at the dorsal edge of the fasciculus longitudinalis medialis; in the golden hamster, mouse, vole, and house shrew, within the central gray matter; and in the guinea pig, at the ventral edge of the fasciculus longitudinalis medialis. In the green monkey and man, we could not find a ventral tegmental nucleus of Gudden, and in the house shrew and man, the pars ventralis and pars dor-salis of the dorsal tegmental nucleus of Gudden could not be separated.

Abstract: Unit discharges associated with ultrasonic vocalization were recorded from the rat medullary region including the nucleus ambiguous (NA). Three types of units were distinguished: ultrasound-related (US), expiration (EXP) and inspiration (INSP) units. Both US and EXP units discharged with tonic bursts mostly prior to ultrasounds. US units were silent during respiration while EXP units discharged synchronously with expiration. During ultrasonic vocalization INSP units suppressed their discharges which were otherwise synchronized with inspiration. Histological examination revealed that US and EXP units were recorded from the ventral division of the NA and the adjacent medullary region where the NA contained large cells and the adjacent region consisted of small cells. These results suggest that both US and EXP units are NA motoneurons responsible for ultrasound emission. EXP units were always supplied with respiratory afferents while US units were not.

Abstract: Unusual paired helical filaments (PHF) coexisting with single filaments were observed in neurofibrillary degeneration (NFD) in the globus pallidus, subthalamic nucleus, substantia nigra, and pontine tegmentum of a typical case of progressive supra-nuclear palsy (PSP). Each filament had a diameter of 10-12 nm and showed central low density and a smooth contour. The thickest portion of a pair was 22-24 nm in diameter. The periodicity of twist varied from 150 nm to 300 nm, but each PHF had regular periodicity. The present ultrastructural finding is unusual in the neurofibrillary pathology of PSP, and is also different from the PHF found in Alzheimer type NFD.

Abstract: Thirty-two patients with hepatocellular carcinoma less than 5 cm in diameter were examined by computed tomography. At the examination 26 patients (81%) were correctly diagnosed. A common type of CT image series was detectable as low density on the precontrast scan, a positive or mixed pattern on dynamic scan, and visible on postcontrast scan. In 10 patients with minute tumors (less than 2 cm), 7 were correctly diagnosed. CT was valuable for diagnosis of the small hepatocellular carcinomas larger than 1 cm by using the dynamic study after an intravenous bolus injection of the contrast medium.

Abstract: Recent progress in clonal cell culture techniques makes it possible to detect pluripotent hemopoietic precursors from murine marrow cells. The precursors can proliferate, differentiate and form mixed colonies containing eryth-roblasts, granulocytes, macrophages and often megakaryocytes in viscid culture medium. In the present investigation, the presence of cells of B-cell lineage in mixed colonies was investigated. Experiments on colonies containing clgM, clgG, slgM and slgG bearing cells using goat IgG fluorescein-conjugated anti-mouse IgM, goat F(ab′)2 fraction fluo-rescein-conjugated anti-mouse IgG and immunobeads revealed the presence of cytoplasmic IgM bearing cells in 47% of the colonies and surface IgM bearing cells in 74-84% of the colonies. Mixed colonies, however, did not contain either clgG bearing cells or slgG bearing cells. The results may indicate that some CFU-MIX proliferate and differentiate along B-cell lineage to slgM or clgM bearing cells in vitro.

Abstract: Seven lymphoblastoid cell lines were established through transformation by Epstein-Barr virus of peripheral blood lymphocytes from two patients with Bloom's syndrome (BS), the parents of a patient, and normal controls. High baseline levels of sister chromatid exchanges (SCEs) in peripheral blood lymphocytes of BS were reduced to about 10% of their initial value in BS lymphoblastoid cell lines, and the elevation of SCE frequencies induced by ethylmethanesulfonate was the same as in controls.

Abstract: The culture for 7days in medium with 5.5mM glucose and 1mM 2-deoxy-Dglucose enhanced the glucose sensitivity of neonatal rat B cells, and even stimulated their growth in vitro. Also, 2-deoxy-D-glucose supplementation maintained insulin release evoked by leucine and 2-ketoisocaproate from B cells at day 7 at levels several times higher than at day 1. The effect of leucine was greatly augmented by glutamine, whereas that of the 2-keto acid remained almost unchanged irrespective of the presence of glutamine. These results suggest an increase in oxidative catabolism of medium nutrients in B cells grown in medium with 2-deoxy-D-glucose for 7days, and such metabolic changes may promote the growth of B cells in vitro.

Abstract: To our knowledge, this is the first report of a patient for whom a successful radical treatment for both membranous obstruction of the inferior vena cava (MOVC) and coincident hepatocellular carcinoma (HCC) were simultaneously carried out. A 36 year old Japanese man with Budd-Chiari syndrome due to MOVC was found to have HCC in the right hepatic lobe during a shunt operation between the inferior vena cava (IVC) and the right atrium (RA), using a vascular prosthetic graft. He was referred to us from another institute for further operative procedures. Both the mediastinum and the abdomen were entered through a long midline incision with longitudinal splitting of the sternum. The graft that had been occluded by thrombus was removed. Transcardiac membranotomy was achieved by manual manipulation and then by Tubb's dilator. The hepatic tumor with an abundant surgical margin of the liver was resected. The tumor of 2.5×2.5×2.0 cm was a well differentiated clear cell type HCC associated with congestive liver cirrhosis. Postoperative contrast and pressure studies through IVC showed satisfactory results. Serum alfa-fetoprotein levels decreased from 503 to 12 ng/ml. The patient is well at least for 44 months after the surgery without any recurrence at the time of completion of this writing.

Abstract: From the present experiments in which sensitization was induced by painting contactants on the same skin of the same mouse over time, the following findings were observed. When mice were sensitized first with PCl, then with DNFB, and finally with OX, the animals showed a normal contact hypersensitivity response to PCl, but abnormal responses to both DNFB and OX, suggesting some suppressor phenomenon. This suppression was shown to be partly due to hapten (DNFB)-specific suppressor cells, and it was suggested that other suppressor cells different from suppressor T cells in nature might be involved. It was considered that induction of suppressor cells might be due to depressed function of the epidermal Langerhans cell affected by potent sensitizers as well as by unknown mechanisms.
