Abstract: 1. The relaxant properties of the type IV adenosine 3',5'-cyclic monophosphate phosphodiesterase (cyclic AMP PDE) inhibitor, rolipram and the beta 2-selective and non-selective beta-adrenoceptor agonists salbutamol and isoprenaline, were compared on the guinea-pig, bovine, and mouse trachea and porcine bronchus all precontracted with methacholine (EC30). 2. Rolipram and both beta-agonists produced concentration-dependent reversal of the methacholine-induced tone in the four airway preparations. 3. Isoprenaline and salbutamol were similar in potency on the guinea-pig (-log10IC50:8.43, 8.06) and bovine (-log10 IC50:8.52, 8.40) airways. In contrast, salbutamol was much less potent than isoprenaline on the mouse trachea (> 1000 fold) and the porcine bronchus (> 100,000 fold). 4. The potency of rolipram approached that of isoprenaline on the guinea-pig and bovine trachea (beta 2-adrenoceptors predominate). However, rolipram was significantly less active than isoprenaline on the porcine bronchus (1000 fold) and mouse trachea (> 2000 fold) where beta 2-adrenoceptors predominate. 5. Siguazodan, the type III cyclic AMP PDE inhibitor, produced concentration-dependent relaxations of the porcine bronchus and guinea-pig trachea contracted with methacholine. Siguazodan was 100 fold more active than rolipram in pig tissues indicating the type III isoenzyme may be of greater functional significance in this tissue. In contrast, siguazodan was 15 times less potent that rolipram in guinea-pig airways suggesting a greater role for the type IV PDE. 6. These findings may reflect a possible relationship between the beta 2-adrenoceptor subtype and the functional importance of the type IV PDE isoenzyme. A similar relationship may exist between beta 1-adrenoceptors and the PDE type III isoenzyme.

Abstract: Novel farnesyl transferase inhibitors of general formula (I), the preparation thereof and pharmaceutical compositions containing said inhibitors, are disclosed. In general formula (I), R1 is Y-S-A1 (where Y is a hydrogen atom, an amino acid residue, a fatty acid residue, an alkyl or alkoxycarbonyl radical or a radical R4-S- where R4 is a C1-4 alkyl radical optionally substituted by a phenyl radical or a radical of general formula (II), wherein A1, X, X1, Y1, R'1, R2, R'2 and R are as defined below, and A1 is a C1-4 alkylene radical optionally alpha-substituted in the ⊃C(X1)(Y1) grouping by an amino or alkylamino, dialkylamino, alkanoylamino or alkoxycarbonylamino radical); X1 and Y1 are each a hydrogen atom or, taken together with the carbon atom to which they are attached, form a ⊃C=O grouping; R'1 is hydrogen or a C1-6 alkyl radical; X is an oxygen or sulphur atom; R2 is a C1-6 alkyl, alkenyl or alkynyl radical optionally substituted by hydroxy, alkoxy, mercapto, alkylthio, alkylsulphinyl or alkylsulphonyl, with the proviso that when R2 is an alkyl radical substituted by a hydroxy radical, R2 may form a lactone with the α-carboxy radical; and R'2 is hydrogen or a C1-6 alkyl radical; and R is a hydrogen atom or an optionally substituted alkyl radical or an optionally substituted phenyl radical, with the proviso that radical (a) is in the 3 or 4 position of the naphthyl ring. The novel products have anticancer properties.

Abstract: A peristaltic pump equipped with at least one peristaltic tube is disclosed characterized in that the said tube is associated with means which limit its occlusion, independently of the physical and/or mechanical stresses to which it may be subjected. The peristaltic tube is provided with at least one longitudinal groove or ridge on the internal surface of its wall. Alternatively, a strand may extend longitudinally inside the peristaltic tube, at least over the part subjected to the action of the pressing devices, and this strand is rigid or semi-rigid and has the shape of an arc of a circle. The strand may be flexible and may be firmly attached to the peristaltic tube at least at a point located upstream from the pressing devices. Various other geometrical arrangements are disclosed. The peristaltic pump is especially adapted to extracorporal blood circulation. The peristaltic tube is of such dimensions that when flattened transversely, there remains at least one passage, the ratio of the cross-section of the passage to the cross-section of the channel of the tube at rest being between 1/10 and 1/10,000. The peristaltic tube consists of a silicone elastomer and may be coated on the internal surface and/or the external surface with a thin layer of a silicone elastomer.

Abstract: Seeds of Pisum sativum contain a biotinyl polypeptide called SBP65 that behaves as a putative sink for the free vitamin, representing more than 90% of the total protein-bound biotin in mature seeds. A cDNA encoding SBP65 was cloned and sequenced. The deduced primary structure of the protein was confirmed by protein sequencing. Peptide sequencing also indicated binding of the biotin to lysine 103. The biotinylation domain of SBP65 differs markedly from that of presently known biotin enzymes. Molecular analysis of the protein sequence reveals an extremely hydrophilic protein containing several repeated motifs. These properties, as well as the temporal and spatial patterns of expression of this protein, suggest that SBP65 belongs to the LEA (late embryogenesis-abundant) group of proteins.
