Abstract: Glucose-grown cells of Bacillus cereus respond to anaerobiosis and low extracellular oxidoreduction potentials (ORP), notably by enhancing enterotoxin production. This response involves the ResDE two-component system. We searched the B. cereus genome for other redox response regulators potentially involved in this adaptive process, and we identified one gene encoding a protein predicted to have an amino acid sequence 58% identical (80% similar) to that of the Bacillus subtilis Fnr redox regulator. The fnr gene of the food-borne pathogen B. cereus F4430/73 has been cloned and partially characterized. We showed that fnr was up-regulated during anaerobic fermentation, especially when fermentation occurred at low ORP (under highly reducing conditions). The expression of fnr was down-regulated in the presence of O(2) and nitrate which, unlike fumarate, stimulated the respiratory pathways. The inactivation of B. cereus fnr abolished fermentative growth but only moderately affected aerobic and anaerobic nitrate respiratory growth. Analyses of glucose by-products and the transcription profiles of key catabolic genes confirmed the strong regulatory impact of Fnr on B. cereus fermentative pathways. More importantly, the fnr mutation strongly decreased the expression of PlcR-dependent hbl and nhe genes, leading to the absence of hemolysin BL (Hbl) and nonhemolytic enterotoxin (Nhe) secretion by the mutant. These data indicate that fnr is essential for both fermentation and toxinogenesis. The results also suggest that both Fnr and the ResDE two-component system belong to a redox regulatory pathway that functions at least partially independently of the pleiotropic virulence gene regulator PlcR to regulate enterotoxin gene expression.

Abstract: Evapotranspiration estimates can be derived from remote sensing data and ancillary, mostly meterorological, information. For this purpose, two types of methods are classically used: the first type estimates a potential evapotranspiration rate from vegetation indices, and adjusts this rate according to water availability derived from either a surface temperature index or a first guess obtained from a rough estimate of the water budget, while the second family of methods relies on the link between the surface temperature and the latent heat flux through the surface energy budget. The latter provides an instantaneous estimate at the time of satellite overpass. In order to compute daily evapotranspiration, one needs an extrapolation algorithm. Since no image is acquired during cloudy conditions, these methods can only be applied during clear sky days. In order to derive seasonal evapotranspiration, one needs an interpolation method. Two combined interpolation/extrapolation methods based on the self preservation of evaporative fraction and the stress factor are compared to reconstruct seasonal evapotranspiration from instantaneous measurements acquired in clear sky conditions. Those measurements are taken from instantaneous latent heat flux from 11 datasets in Southern France and Morocco. Results show that both methods have comparable performances with a clear advantage for the evaporative fraction for datasets with several water stress events. Both interpolation algorithms tend to underestimate evapotranspiration due to the energy limiting conditions that prevail during cloudy days. Taking into account the diurnal variations of the evaporative fraction according to an empirical relationship derived from a previous study improved the performance of the extrapolation algorithm and therefore the retrieval of the seasonal evapotranspiration for all but one datasets.

Abstract: Anthocyanidins are a class of π-conjugated systems responsible for red, blue, and purple colors of plants. They exhibit the capacity of aggregation in the presence of other natural compounds including flavonols. Such complexations induce color modulation in plants, which is known as copigmentation. It is largely driven by π-interactions existing between pigments and copigments. In this work, the energies of copigmentation-complexation and self-association are systematically evaluated for an anthocyanidin/flavonol couple prototype (3-O-methylcyanidin/quercetin). To describe noncovalent interactions, DFT-D appears mandatory to reach a large accuracy. Due to the chemical complexity of this phenomenon, we also aim at assessing the relevance of both B3P86-D2 and ωB97X-D functionals. The benchmarking has shown that B3P86-D2 possesses enough accuracy when dealing with π-π interactions with respect to both spin component scaled Moller-Plesset second-order perturbation theory post Hartree-Fock method and experimental data. UV-vis absorption properties are then evaluated with time-dependent DFT for the different complexes. The use of range-separated hybrid functionals, such as ωB97X-D, helped to correctly disentangle and interpret the origin of the UV-vis experimental shifts attributed to the subtle copigmentation phenomenon.
