Abstract: It is well known that biological motion, as produced by point-light displays on a human body, gives a good representation of the represented body-eg its gender and the nature of the task which it is engaged in. The question is whether it is possible to judge the emotional state of a human body from motion information alone. An ability to make this kind of judgment may imply that people are able to perceive emotion from patterns of movement without having to compute the detailed shape first. Subjects were shown brief video clips of two trained dancers (one male, one female). The dancers were aiming to convey the following emotions: fear, anger, grief, joy, surprise, and disgust. The video clips portrayed fully lit scenes and point-light scenes, with thirteen small points of light attached to the body of each dancer. Half the stimuli were presented the right way up, while half were inverted. The subjects' task was to judge which emotion was being portrayed. Full-body clips gave good recognition of emotionality (88% correct), but the results for upright biological-motion displays were also significantly above chance (63% correct). Inversion of the display reduced biological-motion (but not full-body) performance to close to chance but still significantly above chance. A space-time analysis of the motion of the points of light was carried out, and was related to the discriminability of the different emotions. Biological-motion displays, which convey no information while static, are able to give a rich description of the subject matter, including the ability to judge emotional state. This ability is disrupted when the image is inverted.

Abstract: Vesicular transport to and from the lysosome and late endosome is defective in patients with Chediak-Higashi syndrome (CHS) and in mutant beige (bg) mice. CHS and bg cells have giant, perinuclear vesicles with characteristics of late endosomes and lysosomes that arise from dysregulated homotypic fusion. CHS and bg lysosomes also exhibit compartmental missorting of proteins, such as elastase, glucuronidase and cathepsin G. Lyst, a candidate gene for bg, was identified by direct complementary DNA selection from a yeast artificial chromosome (YAC) clone containing a 650-kilobase segment of the bg-critical region on mouse chromosome 13. Lyst is disrupted by a 5-kilobase deletion in bg mice, and Lyst messenger RNA is markedly reduced in bg homozygotes. The homologous human gene, LYST, is highly conserved with mouse Lyst, and contains a frame-shift mutation at nucleotides 117-118 of the coding domain in a CHS patient. Thus bg mice and human CHS patients have homologous disorders associated with Lyst mutations. Lyst encodes a protein with a carboxy-terminal prenylation motif and multiple potential phosphorylation sites. Lyst protein is predicted to form extended helical domains, and has a region of sequence similar to stathmin, a coiled-coil phosphoprotein thought to act as a relay integrating cellular signal response coupling.

Abstract: The cell-killing effects of the cytokines TNF-alpha and FasL are mediated by the distinct cell-surface receptors TNFR1, TNFR2 and Fas (also known as CD95/APO-1), which are all members of a receptor superfamily that is important for regulating cell survival. The cytoplasmic regions of TNFR1 and Fas contain a conserved 'death' domain which is an essential component of the signal pathway that triggers apoptosis and activation of the transcription factor NF-kappaB (refs 5,6). Here we report the isolation of a 54K receptor that is a new member of the TNFR superfamily, using the death domain of TNFR1 in a yeast two-hybrid system. This protein, WSL-1, is most similar to TNFR1 itself, particularly in the death-domain region. The gene wsl-1 is capable of inducing apoptosis when transfected into 3T3 and 293 cells, and can also activate NF-kappaB in 293 cells. Like TNFR1, WSL-1 will homodimerize in yeast. WSL-1 also interacts specifically with the TNFR1-associated molecule TRADD. The tissue distribution is very restricted and significantly different from that of Fas and TNFR1.

Abstract: Plant cells contain two major pools of K+, one in the vacuole and one in the cytosol. The behavior of K+ concentrations in these pools is fundamental to understanding the way this nutrient affects plant growth. Triple-barreled microelectrodes have been used to obtain the first fully quantitative measurements of the changes in K+ activity (aK) in the vacuole and cytosol of barley (Hordeum vulgare L.) root cells grown in different K+ concentrations. The electrodes incorporate a pH-selective barrel allowing each measurement to be assigned to either the cytosol or vacuole. The measurements revealed that vacuolar aK declined linearly with decreases in tissue K+ concentration, whereas cytosolic aK initially remained constant in both epidermal and cortical cells but then declined at different rates in each cell type. An unexpected finding was that cytoplasmic pH declined in parallel with cytosolic aK, but acidification of the cytosol with butyrate did not reveal any short-term link between these two parameters. These measurements show the very different responses of the vacuolar and cytosolic K+ pools to changes in K+ availability and also show that cytosolic K+ homeostasis differs quantitatively in different cell types. The data have been used in thermodynamic calculations to predict the need for, and likely mechanisms of, active K+ transport into the vacuole and cytosol. The direction of active K+ transport at the vacuolar membrane changes with tissue K+ status.

Abstract: Publisher Summary Soils form a major repository of nitrogen (N) within both natural and agricultural terrestrial ecosystems, containing, on a global basis, an estimated 2.4 x 10 11 tons of N. The soil receives N inputs through fertilizer additions and from the atmosphere in precipitation and dry deposition or via biological fixation; inputs are also made in plant and animal residues. N is removed in the harvested crop and is lost by leaching and surface run-off of soluble forms, by gaseous transfer as N gas and N oxides (during nitrification and denitrification processes), and by ammonia volatilization. In some circumstances, erosion may also be important. In addition to these interactions with the total ecosystem, internal cycles also operate within the soil, so that even if gains and losses are in balance, then N still continues to cycle in the soil. This chapter describes the current understanding of the conceptual basis of the processes involved in mineralization, relationships among the processes and other factors, and how their effects can be determined practically. The aim is to present this in a way that is relevant to current and future agricultural development and to environmental issues.

Abstract: The purpose of this paper is to present an in-depth analysis of the concept of patient participation. The analysis was undertaken using the method described by Walker and Avant. Patient participation is defined and the critical attributes are identified. Model, borderline and contrary cases illustrate what the concept is and is not. The meaning of patient participation is compared with patient partnership, patient collaboration and patient involvement, three related cases. The antecedents and consequences of patient participation are investigated to further refine the critical attributes. Empirical referents are illuminated. The value of concept analysis in relation to precise communication, critical thinking and the advancement of the knowledge base of nursing is discussed.

Abstract: We have cloned two open reading frames (orf6 and orf8) from the Escherichia coli K-12 rfb region. The genes were expressed in E. coli under control of the T7lac promoter, producing large quantities of recombinant protein, most of which accumulated in insoluble inclusion bodies. Sufficient soluble protein was obtained, however, for use in a radiometric assay designed to detect UDP-galactopyranose mutase activity (the conversion of UDP-galactopyranose to UDP-galactofuranose). The assay is based upon high-pressure liquid chromatography separation of sugar phosphates released from both forms of UDP-galactose by phosphodiesterase treatment. The crude orf6 gene product converted UDP-[alpha-D-U-14C]-galactopyranose to a product which upon phosphodiesterase treatment gave a compound with a retention time identical to that of synthetic alpha-galactofuranose-1-phosphate. No mutase activity was detected in extracts from cells lacking the orf6 expression plasmid or from orf8-expressing cells. The orf6 gene product was purified by anion-exchange chromatography and hydrophobic interaction chromatography. Both the crude extract and the purified protein converted 6 to 9% of the UDP-galactopyranose to the furanose form. The enzyme was also shown to catalyze the reverse reaction; in this case an approximately 86% furanose-to-pyranose conversion was observed. These observations strongly suggest that orf6 encodes UDP-galactopyranose mutase (EC 5.4.99.9), and we propose that the gene be designated glf accordingly. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis of purified UDP-galactopyranose mutase revealed one major band, and analysis by electrospray mass spectrometry indicated a single major species with a molecular weight of 42,960 +/- 8, in accordance with that calculated for the Glf protein. N-terminal sequencing revealed that the first 15 amino acids of the recombinant protein corresponded to those expected from the published sequence. UV-visible spectra of purified recombinant enzyme indicated that the protein contains a flavin cofactor, which we have identified as flavin adenine dinucleotide.

Abstract: One of the great strengths of public-key cryptography is its potential to allow the localization of trust. This potential is greatest when cryptography is present to guarantee data integrity rather than secrecy, and where there is no natural hierarchy of trust. Both these conditions are typically fulfilled in the commercial world, where CSCW requires sharing of data and resources across organizational boundaries. One property which trust is frequently assumed or “proved” to have is transitivity (if A trusts B and B trusts C then A trusts C) or some generalization of transitivity such as *-closure. We use the loose term unintensional transitivity of trust to refer to a situation where B can effectively put things into A's set of trust assumptions without A's explicit consent (or sometimes even awareness.) Any account of trust which allows such situations to arise clearly poses major obstacles to the effective confinement (localization) of trust. In this position paper, we argue against the need to accept unintensional transitivity of trust. We distinguish the notion of trust from a number of other (transitive) notions with which it is frequently confused, and argue that “proofs” of the unintensional transitivity of trust typically involve unpalatable logical assumptions as well as undesirable consequences.

Abstract: This paper compares data envelopment analysis (DEA) and ratio analysis as alternative tools for assessing the performance of organisational units such as bank branches and schools. Such units typically use one or more resources to secure one or more outputs, the inputs and/or outputs being possibly incommensurate. The assessment of District Health Authorities in England on the provision of perinatal care is used as a vehicle for comparing the two methods. The comparison focuses on how well the two methods agree on the performance of a unit relative to that of other units, and on the estimates of targets each method provides for improving the performance of units. It is found that provided the performance indicators capture all variables used in the DEA assessment the two methods agree reasonably closely on the performance of the units as a whole, though this depends on the way the performance indicators are combined into a summary figure of performance. The two methods can disagree substantially on the relative performance of individual units. Ratio analysis, unlike DEA, is not found to be suitable for setting targets so that units can become more efficient. This is mainly due to the fact that DEA takes simultaneous account of all resources and outputs in assessing performance while ratio analysis relates only one resource to one output at a time. However, the two methods can support each other if used jointly. Ratios do provide useful information on the performance of a unit on specific aspects and they can support the communication of DEA results to non-specialists when the two methods agree on performance.

Abstract: A novel LDL-associated phospholipase A2 (LDL-PLA2) has been purified to homogeneity from human LDL obtained from plasma apheresis. This enzyme has activity toward both oxidized phosphatidylcholine and platelet activating factor (PAF). A simple purification procedure involving detergent solubilization and affinity and ion exchange chromatography has been devised. Vmax and Km for the purified enzyme are 170 micromol.min-1.mg-1 and 12 micromol/L, respectively. Extensive peptide sequence from LDL-PLA2 facilitated identification of an expressed sequence tag partial cDNA. This has led to cloning and expression of active protein in baculovirus. A lipase motif is also evident from sequence information, indicating that the enzyme is serine dependent. Inhibition by diethyl p-nitrophenyl phosphate and 3,4-dichloroisocoumarin and insensitivity to EDTA, Ca2+, and sulfhydryl reagents confirm that the enzyme is indeed a serine-dependent hydrolase. The protein is extensively glycosylated, and the glycosylation site has been identified. Antibodies to this LDL-PLA2 have been raised and used to show that this enzyme is responsible for >95% of the phospholipase activity associated with LDL. Inhibition of LDL-PLA2 before oxidation of LDL reduces both lysophosphatidylcholine content and monocyte chemoattractant ability of the resulting oxidized LDL. Lysophosphatidylcholine production and monocyte chemoattractant ability can be restored by addition of physiological quantities of pure LDL-PLA2.

Abstract: The most commonly used measures for verifying forecasts or simulators of continuous variables are root-mean-squared error (rmse) and anomaly correlation. Some disadvantages of these measures are demonstrated. Existing assessment systems for categorical forecasts are discussed briefly. An alternative unbiased verification measure is developed, known as the linear error in probability space (LEPS) score. The LEPS scare may be used to assess forecasts of both continuous and categorical variables and has some advantages over rmse and anomaly correlation. The properties of the version of LEPS discussed here are reviewed and compared with an earlier form of LEPS. A skill-score version of LEPS may be used to obtain an overall measure of the skill of a number of forecasts. This skill score is biased, but the bias is negligible if the number of effectively independent forecasts or simulations is large. Some examples are given in which the LEPS skill score is compared with rmse and anomaly correlation.

Abstract: This paper presents the research for the development of a new travel aid to increase the independent mobility of blind and elderly travellers. This aid will build on the technologies of geographical information systems (GIS) and the Global Positioning System (GPS). The MOBIC Travel Aid (MOTA) consists of two interrelated components: the MOBIC Pre-journey System (MOPS) to assist users in planning journeys and the MOBIC Outdoor System (MOODS) to execute these plans by providing users with orientation and navigation assistance during journeys. The MOBIC travel aid is complementary to primary mobility aids such as the long cane or guide dog. Results of a study of user requirements are presented and their implications for the initial design of the system are discussed.

Abstract: 1. Human intestinal epithelial Caco-2 cells have been used to investigate the transepithelial permeation of the cardiac glycoside, digoxin. 2. Transepithelial basal to apical [3H]-digoxin flux exceeds apical to basal flux, a net secretion of [3H]-digoxin being observed. At 200 microM digoxin, net secretory flux (Jnet) was 10.8 +/- 0.6 nmol cm-2 h-1. Maximal secretory flux (Jmax) of vinblastine was 1.3 +/- 0.1 nmol cm-2 h-1. Cellular uptake of digoxin was different across apical and basal cell boundaries. It was greatest across the basal surface at 1 microM, whereas at 200 microM, apical uptake exceeded basal uptake. 3. Net secretion of [3H]-digoxin was subject to inhibition by digitoxin and bufalin but was not inhibited by ouabain, convallatoxin, and strophanthidin (all 100 microM). Inhibition was due to both a decrease in Jb-a and an increase in Ja-b. Uptake of [3H]-digoxin at the apical surface was increased by digitoxin and bufalin. All cardiac glycosides decreased [3H]-digoxin uptake at the basal cell surface (except for 100 microM digitoxin). 4. The competitive P-glycoprotein inhibitors, verapamil (100 microM), nifedipine (50 microM) and vinblastine (50 microM) all abolished net secretion of [3H]-digoxin due to both a decrease in Jb-a and an increase in Ja-b. Cellular accumulation of [3H]-digoxin was also increased across both the apical and basal cell surfaces. I-Chloro-2,4,-dinitrobenzene (10 microM), a substrate for glutathione-S-transferase and subsequent ATP-dependent glutathione-S-conjugate secretion, failed to inhibit net secretion of [3H]-digoxin. The increase in absorptive permeability Pa-b (= Ja-b/Ca) and cellular [3H]-digoxin uptake upon P-glycoprotein inhibition, showed that the intestinal epithelium was rendered effectively impermeable by ATP-dependent extrusion at the apical surface. 5. A model for [3H]-digoxin secretion by the intestinal epithelium is likely to involve both diffusional uptake and Na(+)-K+ pump-mediated endocytosis, followed by active extrusion at the apical membrane.

Abstract: The responses ofPterostichus melanarius Illiger,Harpalus rufipes DeGeer, andNebria brevicollis Fabricius (Coleoptera: Carabidae) to olfactory cues of prey and habitat were studied in a four-arm continuous-airflow olfactometer. The process was semiautomated using time-lapse image analysis by microcomputer. The primary constituent of the aphid alarm pheromone, (E)-β-farnesene (EBF), was synthesized and tested for a kairomonal role in prey detection by carabids. In addition, individual beetles were exposed to odors from live aphids, live collembolans, and a crude extract of wheat. All three beetle species showed evidence of olfaction.P. melanarius responded to all the odors except collembolans,H. rufipes responded to EBF and wheat, andN. brevicollis to collembolans. The use of a defence allomone as a prey-finding kairomone by certain carabid species has implications for pest management. Abstract: The role and functioning of the anterior chemosensory organs of plant parasitic nematodes is examined, with particular emphasis on the amphids. The morphology of the amphids is discussed primarily in the context of the changes in the ultrastructure associated with different life stages. The involvement of amphidial secretions in chemoreception and the behavioral and electrophysiological analyses of nematode responses to semiochemicals are discussed with special reference to research on sex pheromones. These research techniques, combined with the use of lectins and antibodies, provide information on nematode sensilla that may lead to novel control strategies for economically important plant parasitic nematodes based on perturbing nematode sensory perception to prevent host or mate location.

Abstract: Transgenic mice overexpressing the human dysfunctional apolipoprotein E variant, APOE*3 Leiden, develop hyperlipidemia and are highly susceptible to diet-induced atherosclerosis. In the present study, we investigated the effects of diet composition and feeding period on serum cholesterol exposure and the amount of atherosclerosis in the aortic sinus in these mice, using quantitative image analysis. On each of the three diets tested-a low-fat diet, a high-saturated-fat/cholesterol diet, and a high saturated-fat/high- cholesterol/0.5%-cholate diet-transgenic animals showed a marked hyperlipidemia compared with nontransgenic littermates. Measurement of the atherosclerotic lesion areas in cross sections of the aortic sinus in animals exposed to these three diets for up to 6 months showed a 5 to 10 times greater lesion area in transgenic mice compared with nontransgenic controls. Highly significant positive correlations were found between the log-transformed data on lesion area and serum cholesterol exposure (r=.82 to .85 for the 1-, 2-, and 3-month treatment groups), indicating that the hyperlipidemia is likely to be a major determinant in lesion formation. On the basis of these findings, we suggest that the APOE*3 Leiden mouse represents a promising model for intervention studies with hypolipidemic and antiatherosclerotic drugs. Abstract: A combined sensor strip was fabricated by screen-printing a Meldola Blue modified carbon working electrode and a Ag/AgCl combined reference/counter electrode alongside each other on a PVC substrate. This was investigated as an amperometric sensor for NADH, at an applied potential of 0 V, both in bulk solutions under hydrodynamic conditions and using 20 μL volumes without agitation. The former technique gave linear responses in the range 3–60 × 10−6mol dm−3 and the latter in the range 10−5 × 10−4 mol dm−3 of NADH. Further modification of the electrodes with NAD+ and lactate dehydrogenase converted the device to a biosensor for lactate both in spiked phosphate buffer and bovine serum. The sensors demonstrated good linear responses in the range 0–20 × 10−3 mol dm−3.
