Position-dependent effects on stability in tricyclo-DNA modified oligonucleotide duplexes

TL;DR: CD-spectroscopic investigations in combination with molecular dynamics simulations of duplexes with single modifications show that tc-residues in the RNA backbone distinctly influence the conformation of the neighboring nucleotides forcing them into higher energy conformations, while tc-Residue in the DNA backbone seem to have negligible influence on the nearest neighbor conformations.

TL;DR: It was found that different C8' substituents (at the bottom of the minor groove) on carba-ENA-T only led to rather small variation of thermal stability of the AON:RNA duplexes, but it was observed that the parent carBA-ENA -T modified AONs exhibited higher nucleolytic stability than those of the ENA-T modified counterparts.

Kinetics of DNA Strand Displacement Systems with Locked Nucleic Acids B

TL;DR: In this article, the authors studied the strand displacement kinetics as a function of the number and position of LNA nucleotides in DNA oligonucleotides and found that the rational substitution of LNAs for DNA nucleotide preserves sequence space while improving the signal-to-noise ratio of strand displacement systems.
