Abstract
Here, we describe the third major release of RELION CPU-based vector acceleration has been added in addition to GPU support, which provides flexibility in use of resources and avoids memory limitations Reference-free autopicking with Laplacian-of-Gaussian filtering and execution of jobs from python allows non-interactive processing during acquisition, including 2D-classification, de novo model generation and 3D-classification Per-particle refinement of CTF parameters and correction of estimated beam tilt provides higher-resolution reconstructions when particles are at different heights in the ice, and/or coma-free alignment has not been optimal Ewald sphere curvature correction improves resolution for large particles We illustrate these developments with publicly available data sets: together with a Bayesian approach to beam-induced motion correction it leads to resolution improvements of 02-07 A compared to previous RELION versions

Real-time cryo-electron microscopy data preprocessing with Warp.

TL;DR: Warp is described, a software that automates all preprocessing steps of cryo-EM data acquisition and enables real-time evaluation, and includes deep-learning-based models for accurate particle picking and image denoising.

Multifunctional graphene supports for electron cryomicroscopy.

TL;DR: A multifunctional specimen support for cryoEM is presented, comprising large-crystal monolayer graphene suspended across the surface of an ultrastable gold specimen support that reduces specimen movement during imaging, improves image quality, and allows high-resolution structure determination with a minimum of material and data.

Heparin-induced tau filaments are polymorphic and differ from those in Alzheimer's and Pick's disease

TL;DR: The results indicate that tau filaments are structurally versatile, and raise questions about the relevance of in vitro assembled amyloids, which have larger cores with different repeat compositions.

Cryo-EM structure of the homohexameric T3SS ATPase-central stalk complex reveals rotary ATPase-like asymmetry.

TL;DR: In this paper, the authors presented the 3.3-dimensional structure of the E.coli T3SS ATPase-central stalk complex, which formed a homohexameric, asymmetric pore with different functional states.

Structure of the gene therapy vector, adeno-associated virus with its cell receptor, AAVR

TL;DR: AAVR’s footprint overlaps the epitopes of several neutralizing antibodies, prompting a re-evaluation of neutralization mechanisms, and the structure provides a roadmap for experimental probing and manipulation of viral-receptor interactions.

A simplex method for function minimization

TL;DR: A method is described for the minimization of a function of n variables, which depends on the comparison of function values at the (n 41) vertices of a general simplex, followed by the replacement of the vertex with the highest value by another point.

RELION: implementation of a Bayesian approach to cryo-EM structure determination.

TL;DR: Developments that reduce the computational costs of the underlying maximum a posteriori (MAP) algorithm, as well as statistical considerations that yield new insights into the accuracy with which the relative orientations of individual particles may be determined are described.

cryoSPARC: algorithms for rapid unsupervised cryo-EM structure determination

TL;DR: It is shown that stochastic gradient descent (SGD) and branch-and-bound maximum likelihood optimization algorithms permit the major steps in cryo-EM structure determination to be performed in hours or minutes on an inexpensive desktop computer.

Automated electron microscope tomography using robust prediction of specimen movements.

TL;DR: A new method was developed to acquire images automatically at a series of specimen tilts, as required for tomographic reconstruction, using changes in specimen position at previous tilt angles to predict the position at the current tilt angle.
