Abstract: Oils and fats are frozen two times from acetone solution at -80 C under protection by ascorbyl palmitate. Tocopherols (T) and tocotrienols (T3) present in the filtered extract are separated into their homologues by one-dimensional thin layer chromatography on precoated silica gel plates in the n-hexane/ethyl acetate system 92.5:7.5. Complete separation of the positional isomers β-T and γ-T is accomplished as well, whereas β-T3 and γ-T are assumed to form identical bands. Suitable spray- and detection systems including new found coloring ones are described for the qualitative estimation of the chromatograms. The content of individual tocopherols (T and T3) of 24 commercial vegetable oils is quantitatively determined using the Emmerie-Engel procedure.

Abstract: A method has been developed for the determination of perazine, clozapine, imipramine and amitriptyline and their demethylated metabolites in plasma. Other metabolites measured were perazine sulfoxide and the N-oxides of clozapine and perazine, the latter two following their reduction to the parent drugs with ascorbic acid. 10-Hydroxynortriptyline was identified as an amitriptyline metabolite in plasma. The general procedure included extraction of alkalinized plasma samples (3 – 6 g) with benzene or toluene and thin layer chromatography of the extracts, followed by reflectance photometry of the plates at appropriate wave lengths in ultraviolet light. Spots of questionable identity were further characterized by two-dimensional chromatography and by colour reactions. The recoveries of compounds added in therapeutic concentrations were between 70 and 98 %. The limits of detectability were 5 – 10 ng/g plasma.

Abstract: Calcitonin was extracted from the pericardium and esophagus of eel in quantities sufficient to permit purification and chemical characterization. Homogeneous calcitonin could be isolated by a six-step fractionation starting from acetone powder of the organs. The fractionation procedure consisted of acid extraction, gel filtration on Sephadex G-75, chromatography on SP-Sephadex C-25, gel filtration on the Sephadex G-50, chromatography on carboxymethylcellulose, and gel filtration on Sephadex G-50. Fractionation of the hormone was monitored by assay of its biological activity and from its behaviour on thin layer chromatography and polyacrylamide gel disc electrophoresis. The hormone contained 32 amino acid residues, like calcitonins from other species of animals, but its amino acid composition was different from those of previously characterized hormones. Eel calcitonin possessed almost the same, or higher, biological activity as the salmon or chicken hormone, which show the highest specific activity among calcitonins so far isolated.

Abstract: The contents of phosphatidyl N-methyl-and N,N-dimethylaminoethanol were determined in the liver of rats injected with (Me-C14) methionine. Total phospholipids were extracted from aliquots of the liver and fractionated by two-dimensional thin layer chromatography after addition of carrier phosphatidyl-N-methyl-and N,N-dimethylaminoethanol. The radioactivity present in the two phosphatide spots was determined and used to calculated total disintegrations per min/100 g body wt. The remainder of the livers was pooled, and total phospholipids were isolated and subjected to acid hydrolysis. N-methyl- and N,N-dimethylaminoethanol were purified by thin layer chromatography, and their specific activity was determined after quantitation by gas liquid chromatography and radioactivity measurement. The liver contents of phosphatidyl N-methyl- and N,N-dimethylaminoethanol were determined by dividing disintegrations per min/100 g body wt by the specific activity of N-methyl- or N,N-dimethylaminoethanol.

Abstract: アミノ酸およびアマイドをシリカゲル薄層クロマトグラフィにより分離し, その重窒素濃度を発光分光分析法により測定する方法についていくつかの検討を行った。その結果シリカゲルからの窒素の汚染が大きいために, シリカゲルをそのまま封電管内に封入する従来法による測定では, かなりの誤差を生じることが明らかになった。この汚染を回避するために, シリカゲルを560℃で30分焼き, さらに分離, 採取されたシリカゲル中のアミノ酸およびアマイドを50%エタノールで溶出し, 封電管中にはシリカゲルを含まない方法を考案した。これにより窒素の汚染量は従来法の1/5程度まで減少した。この方法を植物より分離されたアミノ酸およびアマイドの重窒素濃度の測定に適用した結果, 従来法よりもすぐれていることが明らかにされた。

Abstract: Methods of measuring the vibrational spectra of species adsorbed on thin layer chromatography (TLC) plates have been studied by Fourier transform infrared (FT-ir) and Raman spectrometry. Previous measurements of the transmittance spectra of species separated by TLC (with the adsorbent deposited on AgCl plates) showed poor signal/noise ratio at high frequency due to scattering by the adsorbent. Scattering has been minimized in these experiments by treating the TLC plate with Fluorolube, an infrared mulling oil whose refractive index closely matches that of the adsorbent. Spectra of chlorinated hydrocarbon pesticides on silica gel and alumina adsorbents measured in this way show differences which can be correlated with the mechanism of adsorption. FT-ir spectra of compounds on TLC plates can be obtained from microgram quantities of sample but Raman spectrometry is not as sensitive, requiring at least 200 μg of sample. The Raman bands of adsorbed pesticides are weak and considerably broadened with respect to the corresponding bands in the spectra of the pure samples.
