HDAC6 inhibition reverses axonal transport defects in motor neurons derived from FUS-ALS patients
Wenting Guo,Maximilian Naujock,Maximilian Naujock,Laura Fumagalli,Tijs Vandoorne,Pieter Baatsen,Ruben Boon,Laura Ordovás,Laura Ordovás,Abdulsamie Patel,Marc Welters,Thomas Vanwelden,Natasja Geens,Tine Tricot,Veronick Benoy,Jolien Steyaert,Cynthia Lefebvre-Omar,Werend Boesmans,Matthew Jarpe,Jared Sterneckert,Florian Wegner,Susanne Petri,Delphine Bohl,Pieter Vanden Berghe,Wim Robberecht,Philip Van Damme,Catherine M. Verfaillie,Ludo Van Den Bosch +27 more
TL;DR: Using motor neurons derived from induced pluripotent stem cells from patients with ALS and FUS mutations, the authors demonstrate that axonal transport deficits that are observed in these cells can be rescued by HDAC6 inhibition.
Journal ArticleDOI
Mitochondrial defect in muscle precedes neuromuscular junction degeneration and motor neuron death in CHCHD10 S59L/+ mouse.
Emmanuelle Génin,Blandine Madji Hounoum,Sylvie Bannwarth,Konstantina Fragaki,Sandra Lacas-Gervais,Alessandra Mauri-Crouzet,Françoise Lespinasse,Julien Neveu,Baptiste Ropert,Gaëlle Augé,Charlotte Cochaud,Cynthia Lefebvre-Omar,Stéphanie Bigou,Aude Chiot,Fanny Mochel,Séverine Boillée,Christian S. Lobsiger,Delphine Bohl,Jean-Ehrland Ricci,Véronique Paquis-Flucklinger +19 more
TL;DR: The data confirm that mitochondrial deficiency associated with CHCHD10 mutations can be at the origin of MND and show that the pathological effects of the p.Ser59Leu mutation target muscle prior to NMJ and motor neurons.
Journal ArticleDOI
Structure-based optimization of a PDZ-binding motif within a viral peptide stimulates neurite outgrowth
Zakir Khan,Elouan Terrien,Florent Delhommel,Cynthia Lefebvre-Omar,Delphine Bohl,Sandrine Vitry,Clara Bernard,Juan Ramirez,Alain Chaffotte,Kevin Ricquier,Renaud Vincentelli,Henri Buc,Christophe Prehaud,Nicolas Wolff,Monique Lafon +14 more
TL;DR: Findings indicate that virus-derived peptides targeted to MAST2–PDZ stimulate neurite outgrowth in several neuron types, opening up promising avenues for potentially using NVs in the management of neurodegenerative diseases.
Journal ArticleDOI
Neurofilament accumulations in amyotrophic lateral sclerosis patients’ motor neurons impair axonal initial segment integrity
Cynthia Lefebvre-Omar,Elise Liu,Carine Dalle,Boris Lamotte d'Incamps,Stéphanie Bigou,Clement Daube,Léa Karpf,Marc Davenne,Noémie Robil,Stéphane Blanchard,Guillaume Tournaire,Charles Nicaise,François Salachas,Lucette Lacomblez,Danielle Seilhean,Christian S. Lobsiger,Stéphanie Millecamps,Séverine Boillée,Delphine Bohl +18 more
TL;DR: In this paper , the authors showed that light NF (NF-L) chains rapidly accumulate in MN soma, while the phosphorylated heavy/medium NF (pNF-M/H) chains pile up in axonal proximal regions of only C9orf72 and SOD1 MNs.
