HDAC6 inhibition reverses axonal transport defects in motor neurons derived from FUS-ALS patients
Wenting Guo,Maximilian Naujock,Maximilian Naujock,Laura Fumagalli,Tijs Vandoorne,Pieter Baatsen,Ruben Boon,Laura Ordovás,Laura Ordovás,Abdulsamie Patel,Marc Welters,Thomas Vanwelden,Natasja Geens,Tine Tricot,Veronick Benoy,Jolien Steyaert,Cynthia Lefebvre-Omar,Werend Boesmans,Matthew Jarpe,Jared Sterneckert,Florian Wegner,Susanne Petri,Delphine Bohl,Pieter Vanden Berghe,Wim Robberecht,Philip Van Damme,Catherine M. Verfaillie,Ludo Van Den Bosch +27 more
TL;DR: Using motor neurons derived from induced pluripotent stem cells from patients with ALS and FUS mutations, the authors demonstrate that axonal transport deficits that are observed in these cells can be rescued by HDAC6 inhibition.
Journal ArticleDOI
Amino Acid Levels Determine Metabolism and CYP450 Function of Hepatocytes and Hepatoma Cell Lines
Ruben Boon,Manoj Kumar,Tine Tricot,Ilaria Elia,Laura Ordovás,Laura Ordovás,Frank Jacobs,Jennifer One,Jonathan De Smedt,Guy Eelen,Matthew Bird,Philip Roelandt,Ginevra Doglioni,Kim Vriens,Matteo Rossi,Marta Aguirre Vazquez,Thomas Vanwelden,Francois Chesnais,Adil El Taghdouini,Mustapha Najimi,Etienne Sokal,David Cassiman,Jan Snoeys,Mario Monshouwer,Wei Shou Hu,Christian Lange,Peter Carmeliet,Sarah-Maria Fendt,Catherine M. Verfaillie +28 more
TL;DR: It is demonstrated that increasing extracellular amino acids beyond the nutritional need of HLCs and HepG2 cells induces glucose independence, mitochondrial function, and the acquisition of a transcriptional profile that is closer to PHHs.
Journal ArticleDOI
Highly proliferative primitive fetal liver hematopoietic stem cells are fueled by oxidative metabolic pathways.
Javed Karim Manesia,Zhuofei Xu,Dorien Broekaert,Ruben Boon,Alexander R. van Vliet,Guy Eelen,Thomas Vanwelden,Steve Stegen,Nick van Gastel,Alberto Pascual-Montano,Sarah-Maria Fendt,Geert Carmeliet,Peter Carmeliet,Satish Khurana,Catherine M. Verfaillie +14 more
TL;DR: It is reported that FL HSCs use oxygen dependent energy generating pathways significantly more than their BM counterparts, and that higher levels of DNA repair and antioxidant pathway gene expression may prevent ROS-mediated (geno)toxicity in FL H SCs.
Journal ArticleDOI
Prdm12 Directs Nociceptive Sensory Neuron Development by Regulating the Expression of the NGF Receptor TrkA
Simon Desiderio,Simon Vermeiren,Claude Van Campenhout,Sadia Kricha,Elisa Malki,Elisa Malki,Sven Richts,Emily V. Fletcher,Thomas Vanwelden,Béla Z. Schmidt,Kristine A. Henningfeld,Tomas Pieler,C. Geoffrey Woods,Vanja Nagy,Catherine M. Verfaillie,Eric Bellefroid +15 more
TL;DR: It is shown that Prdm12 expression is restricted to developing and adult nociceptors and that its genetic ablation compromises their viability and maturation.
Journal ArticleDOI
Distinct Molecular Signature of Murine Fetal Liver and Adult Hematopoietic Stem Cells Identify Novel Regulators of Hematopoietic Stem Cell Function
Javed Karim Manesia,Mónica Franch,Daniel Tabas-Madrid,Rubén Nogales-Cadenas,Thomas Vanwelden,Elisa Van Den Bosch,Zhuofei Xu,Alberto Pascual-Montano,Satish Khurana,Satish Khurana,Catherine M. Verfaillie +10 more
TL;DR: In this paper, RNA sequencing-based gene expression analysis on 14.5-day mouse embryo (E14.5) was performed to identify key transcriptional regulators that play important roles in the expansion of HSCs during development.
