TL;DR: Genetic studies in Drosophila suggest that changes in cellular pyruvate metabolism are necessary and sufficient to promote cancer initiation, and loss of Mpc1 prior to a tumorigenic stimulus doubled the frequency of adenoma formation and produced higher grade tumors.

TL;DR: An important role is defined for cellular pyruvate metabolism in differentiated enterocytes to maintain stem cell proliferation rates and upd3 and JNK signaling are required in enterocytes for ISC proliferation, indicating that this reflects a canonical non-cell autonomous damage response.
