Journal ArticleDOI
TNF-α–driven inflammation and mitochondrial dysfunction define the platelet hyperreactivity of aging
Pavel Davizon-Castillo,Brandon McMahon,Sonia Aguila,David Bark,Katrina J. Ashworth,Ayed Allawzi,Robert A. Campbell,Emilie Montenont,Travis Nemkov,Angelo D'Alessandro,Nathan Clendenen,Lauren Shih,Natalie A. Sanders,Natalie A. Sanders,Kelly Higa,Allaura Cox,Zavelia Padilla-Romo,Giovanni Hernandez,Eric P. Wartchow,George Devon Trahan,Eva Nozik-Grayck,Kenneth L. Jones,Eric M. Pietras,James DeGregori,Matthew T. Rondina,Jorge Di Paola +25 more
TL;DR: Previously-unrecognized evidence is presented that TNF- critically regulates megakaryocytes resident in the bone marrow niche and aging-associated platelet hyperreactivity and thrombosis and age-dependent metabolic profiles that may differentially poise platelets for activation.
Journal ArticleDOI
Neutrophil-derived S100 calcium-binding proteins A8/A9 promote reticulated thrombocytosis and atherogenesis in diabetes
Michael J Kraakman,Man K.S. Lee,Annas Al-Sharea,Dragana Dragoljevic,Tessa J. Barrett,Emilie Montenont,Debapriya Basu,Sarah E Heywood,Helene L. Kammoun,Michelle C Flynn,Alexandra Whillas,Nordin M J Hanssen,Nordin M J Hanssen,Mark A. Febbraio,Erik Westein,Edward A. Fisher,Jaye Chin-Dusting,Mark E. Cooper,Jeffrey S. Berger,Ira J. Goldberg,Prabhakara R Nagareddy,Andrew J. Murphy,Andrew J. Murphy +22 more
TL;DR: In response to hyperglycemia, neutrophil-derived S100 calcium-binding proteins A8/A9 (S100A8/ a9) interact with the receptor for advanced glycation end products (RAGE) on hepatic Kupffer cells, resulting in increased production of IL-6, a pleiotropic cytokine that is implicated in inflammatory thrombocytosis.
Journal ArticleDOI
Aspirin attenuates platelet activation and immune activation in HIV-1-infected subjects on antiretroviral therapy: a pilot study.
Meagan O’Brien,Emilie Montenont,Liang Hu,Michael A. Nardi,Vanessa Valdes,Michael Merolla,Gabrielle Gettenberg,Karen Cavanagh,Judith A. Aberg,Nina Bhardwaj,Jeffrey S. Berger +10 more
TL;DR: It is demonstrated that heightened platelet activation and immune activation in treated HIV-1 disease are attenuated by 1 week of aspirin therapy.
Journal ArticleDOI
Human megakaryocytes possess intrinsic antiviral immunity through regulated induction of IFITM3.
Robert A. Campbell,Hansjörg Schwertz,Eugenio D. Hottz,Eugenio D. Hottz,Jesse W. Rowley,Bhanu Kanth Manne,A. Valance Washington,A. Valance Washington,Robert F. Hunter-Mellado,Robert F. Hunter-Mellado,Neal D. Tolley,Miles Christensen,Alicia S. Eustes,Emilie Montenont,Seema Bhatlekar,Cassandra H. Ventrone,Beth D. Kirkpatrick,Kristen K. Pierce,Stephen S. Whitehead,Sean A. Diehl,Paul F. Bray,Guy A. Zimmerman,Yasuhiro Kosaka,Patrícia T. Bozza,Fernando A. Bozza,Andrew S. Weyrich,Matthew T. Rondina,Matthew T. Rondina +27 more
TL;DR: It is established, for the first time, that human MKs possess antiviral functions, preventing DENV infection of MKs and hematopoietic stem cells after local immune signaling.
Journal ArticleDOI
Rap1 and its effector RIAM are required for lymphocyte trafficking.
Wenjuan Su,Joseph Wynne,Elaine M. Pinheiro,Marianne Strazza,Adam Mor,Emilie Montenont,Jeffrey S. Berger,David S. Paul,Wolfgang Bergmeier,Frank B. Gertler,Mark R. Philips +10 more
TL;DR: In vivo results confirm a role for RIAM in the regulation of some, but not all, leukocyte integrins and suggest that RIAM-regulated integrin activation is required for trafficking of lymphocytes from blood into pLNs and BM, where relatively high shear forces exist in high endothelial venules and sinusoids, respectively.
