Mutant KRAS -driven cancers depend on PTPN11 /SHP2 phosphatase
Dietrich A. Ruess,Dietrich A. Ruess,Guus J.J.E. Heynen,Katrin J. Ciecielski,Jiaoyu Ai,Alexandra Berninger,Derya Kabacaoglu,Kivanc Görgülü,Zahra Dantes,S Wörmann,Kalliope N. Diakopoulos,Angeliki Faidra Karpathaki,Marlena Kowalska,Ezgi Kaya-Aksoy,Liang Song,Eveline A.N. Zeeuw van der Laan,María P. López-Alberca,Marc Nazaré,Maximilian Reichert,Dieter Saur,Mert Erkan,Ulrich T. Hopt,Bruno Sainz,Walter Birchmeier,Roland M. Schmid,Marina Lesina,Hana Algül +26 more
TL;DR: The data indicate the clinical utility of dual SHP2/MEK inhibition as a targeted therapy approach for KRAS-mutant cancers and provide evidence for a critical dependence of mutant KRAS on SHp2 during carcinogenesis.
Journal ArticleDOI
Splicing modulation as novel therapeutic strategy against diffuse malignant peritoneal mesothelioma.
Rocco Sciarrillo,Anna Wojtuszkiewicz,Btissame El Hassouni,Niccola Funel,Paolo Gandellini,Tonny Lagerweij,Silvia Buonamici,Maxime Blijlevens,Eveline A.N. Zeeuw van der Laan,Nadia Zaffaroni,Marcello Deraco,Shigeki Kusamura,Thomas Wurdinger,Godefridus J. Peters,Carla F. M. Molthoff,Gerrit Jansen,Gertjan J.L. Kaspers,Jacqueline Cloos,Elisa Giovannetti,Elisa Giovannetti +19 more
TL;DR: Splicing modulators markedly impair cancer cell viability, resulting also in potent antitumor activity in vivo, and this data designate splicing as a promising therapeutic target in DMPM.
Journal ArticleDOI
Impact of hypoxia on chemoresistance of mesothelioma mediated by the proton-coupled folate transporter, and preclinical activity of new anti-LDH-A compounds
Giovanna Li Petri,Giovanna Li Petri,Btissame El Hassouni,Rocco Sciarrillo,Niccola Funel,Giulia Mantini,Eveline A.N. Zeeuw van der Laan,Stella Cascioferro,Amir Avan,Amir Avan,Paolo Andrea Zucali,Nadia Zaffaroni,Tonny Lagerweij,Barbara Parrino,Kees Smid,Marcello Deraco,Carlotta Granchi,Alicja Braczko,Ryszard T. Smolenski,Larry H. Matherly,Gerrit Jansen,Yehuda G. Assaraf,Patrizia Diana,Jacqueline Cloos,Godefridus J. Peters,Godefridus J. Peters,Filippo Minutolo,Elisa Giovannetti +27 more
TL;DR: Novel insights into hypoxia/PCFT-dependent chemoresistance are provided, unravelling the potential prognostic value of LDH-A, and demonstrating the preclinical activity of LDh-A inhibitors are demonstrated.
Journal ArticleDOI
Update on the pathophysiology of transfusion-related acute lung injury.
Eveline A.N. Zeeuw van der Laan,Saskia van der Velden,Leendert Porcelijn,John W. Semple,C. Ellen van der Schoot,Rick Kapur +5 more
TL;DR: Apart from PMNs, macrophages also appear to be important in the pathogenesis of TRALI, and targeting the pathogenic functions of macrophage may be a promising therapeutic strategy to explore in TRALi.
Journal ArticleDOI
Evaluation of Platelet Responses in Transfusion-Related Acute Lung Injury (TRALI)
Eveline A.N. Zeeuw van der Laan,Saskia van der Velden,Leendert Porcelijn,John W. Semple,C. Ellen van der Schoot,Rick Kapur +5 more
TL;DR: It appears that recipient platelets may play a pathogenic role in antibody-mediated TRALI, however, their contribution appears to be limited and it is imperative to first validate the involvement of recipient platelet by standardizing the animal models, methods, reagents and read-outs for lung injury and taking the animal housing environment into consideration.
