A Novel Progranulin Mutation Causing Frontotemporal Lobar Degeneration with Heterogeneous Phenotypic Expression
Giacomina Rossi,Elena Piccoli,Luisa Benussi,Francesca Caso,Veronica Redaelli,Giuseppe Magnani,Giuliano Binetti,Roberta Ghidoni,Daniela Perani,Giorgio Giaccone,Fabrizio Tagliavini +10 more
TL;DR: An Italian family with FTLD showing remarkable phenotypic heterogeneity is described, with low plasma levels of progranulin, and the novel frame-shift mutation T278SfsX7 found in two patients with early onset is analyzed.
Journal ArticleDOI
Ubiquitinated neurites are associated with preamyloid and cerebral amyloid β deposits in patients with hereditary cerebral hemorrhage with amyloidosis Dutch type
TL;DR: The study showed that amyloid deposits in the vessel walls and in the neuropil were surrounded by abnormal ubiquitinated neurites, suggesting that Aβ deposition induces neuritic changes.
Journal ArticleDOI
Faster Cortical Thinning and Surface Area Loss in Presymptomatic and Symptomatic C9orf72 Repeat Expansion Adult Carriers.
Gabriella Le Blanc,Vincent Jetté Pomerleau,Jillian McCarthy,Barbara Borroni,John C. van Swieten,Daniela Galimberti,Raquel Sánchez-Valle,Robert Laforce,Fermin Moreno,Matthis Synofzik,Caroline Graff,Mario Masellis,Maria Carmela Tartaglia,James B. Rowe,Rik Vandenberghe,Elizabeth Finger,Fabrizio Tagliavini,Alexandre de Mendonça,Isabel Santana,Christopher C Butler,Alexander Gerhard,Adrian Danek,Adrian Danek,Johannes Levin,Markus Otto,Giovanni B. Frisoni,Sandro Sorbi,Jonathan D. Rohrer,Simon Ducharme,Simon Ducharme +29 more
TL;DR: Ageing trajectories of cortical thickness and surface area in C9orf72 expansion adult carriers compared to healthy controls are examined to characterize preclinical cerebral changes leading to symptoms of frontotemporal dementia.
Journal ArticleDOI
Dreaming of a New World Where Alzheimer's Is a Treatable Disorder.
TL;DR: mounting evidence from scientific literature supports the view of AD as a multifactorial disease that results from the concomitant action of multiple molecular players and suggests that AD drug design needs to be shifted towards multi-targeted compounds or drug combinations acting synergistically on the main core features of disease pathogenesis.
Journal ArticleDOI
Ventricular volume expansion in presymptomatic genetic frontotemporal dementia
Tamara P. Tavares,Derek G.V. Mitchell,Kristy Coleman,Christen Shoesmith,Robert Bartha,David M. Cash,Katrina M. Moore,John C van Swieten,Barbara Borroni,Daniela Galimberti,Maria Carmela Tartaglia,James B. Rowe,Caroline Graff,Fabrizio Tagliavini,Giovanni B. Frisoni,Stefano F. Cappa,Robert Laforce,Alexandre de Mendonça,Sandro Sorbi,Garrick Wallstrom,Mario Masellis,Jonathan D. Rohrer,Elizabeth Finger +22 more
TL;DR: In this article, the authors used linear mixed models to identify differences in ventricular volume and in expansion rates as a function of time to expected disease onset between presymptomatic carriers and noncarriers.
