Journal Article
Biochemical and pharmacological characterization of the cyclooxygenase activity of human blood prostaglandin endoperoxide synthases.
Paola Patrignani,M. R. Panara,Anita Greco,Ornella Fusco,Clara Natoli,Stefano Iacobelli,Francesco Cipollone,A. Ganci,C Créminon,Jacques Maclouf +9 more
TL;DR: A model of human prostaglandin endoperoxide synthase-2 expression allowing the assessment of pharmacological inhibition in vitro and ex vivo was characterized and cyclooxygenase activity increased in parallel with the mass of a monocyte protein doublet analyzed by Western blot using antibodies directed against the carboxyl-terminal portion of human PGHS-2.
Journal ArticleDOI
The receptor RAGE as a progression factor amplifying arachidonate-dependent inflammatory and proteolytic response in human atherosclerotic plaques: role of glycemic control.
Francesco Cipollone,Annalisa Iezzi,Maria Fazia,Mirco Zucchelli,Barbara Pini,Chiara Cuccurullo,Domenico De Cesare,Giovanni De Blasis,Raffaella Muraro,Roberto Bei,Francesco Chiarelli,Ann Marie Schmidt,Franco Cuccurullo,Andrea Mezzetti +13 more
TL;DR: It is demonstrated in humans that RAGE overexpression is associated with enhanced inflammatory reaction and COX-2/mPGES-1 expression in diabetic plaque macrophages, and this effect may contribute to plaque destabilization by inducing culprit metalloproteinase expression.
Journal ArticleDOI
Overexpression of functionally coupled cyclooxygenase-2 and prostaglandin E synthase in symptomatic atherosclerotic plaques as a basis of prostaglandin E(2)-dependent plaque instability.
Francesco Cipollone,Cesaria Prontera,Barbara Pini,Matteo Marini,Maria Fazia,Domenico De Cesare,Annalisa Iezzi,Sante Ucchino,Gianfranco Boccoli,Vittorio Saba,Francesco Chiarelli,Franco Cuccurullo,Andrea Mezzetti +12 more
TL;DR: This study provides evidence that synthesis of COX-2 and PGES by activated macrophages is associated with acute ischemic syndromes, possibly through metalloproteinase-induced plaque rupture.
Journal ArticleDOI
In Vivo Formation of 8-Epi-Prostaglandin F2α Is Increased in Hypercholesterolemia
Giovanni Davì,Paola Alessandrini,Andrea Mezzetti,Giorgio Minotti,Tonino Bucciarelli,Fabrizio Costantini,Francesco Cipollone,Gabriele Bittolo Bon,Giovanni Ciabattoni,Carlo Patrono +9 more
TL;DR: It is concluded that the in vivo formation of the F2-isoprostane 8-epi-PGF2α is enhanced in the vast majority of patients with hypercholesterolemia and dose-dependent suppression of enhanced 8-Epi- PGF2 α formation by vitamin E supplementation may contribute to the beneficial effects of antioxidant treatment.
Journal ArticleDOI
A polymorphism in the cyclooxygenase 2 gene as an inherited protective factor against myocardial infarction and stroke.
Francesco Cipollone,Elena Toniato,Stefano Martinotti,Maria Fazia,Annalisa Iezzi,Chiara Cuccurullo,Barbara Pini,Sebastiano Ursi,Gianfranco Vitullo,Maurizio Averna,Marcello Arca,Anna Montali,Filomena Campagna,Sante Ucchino,Francesco Spigonardo,Stefano Taddei,Agostino Virdis,Giovanni Ciabattoni,Alberto Notarbartolo,Franco Cuccurullo,Andrea Mezzetti +20 more
TL;DR: It is found that the -765G-->C polymorphism of the COX-2 gene is associated with a decreased risk of MI and stroke and detection of this genotype may be useful for predicting genetic risk ofMI and stroke.
