Journal ArticleDOI
Targeted Overexpression of Transmembrane Tumor Necrosis Factor Provokes a Concentric Cardiac Hypertrophic Phenotype
Ziad Dibbs,Abhinav Diwan,Shintaro Nemoto,Gilberto DeFreitas,Maha Abdellatif,Blase A. Carabello,Francis G. Spinale,Giora Z. Feuerstein,Natarajan Sivasubramanian,Douglas L. Mann +9 more
TL;DR: These studies show that transmembrane TNF is biologically active and provokes a concentric hypertrophic cardiac phenotype, thus suggesting that posttranslational processing (ie, secretion) of T NF is responsible for the dilated cardiomyopathic phenotype in mice with targeted, cardiac-restricted overexpression of TNF.
Journal ArticleDOI
Cardiac Restricted Overexpression of Membrane Type-1 Matrix Metalloproteinase Causes Adverse Myocardial Remodeling following Myocardial Infarction
Francis G. Spinale,Rupak Mukherjee,Juozas A. Zavadzkas,Christine N. Koval,Shenikqua Bouges,Robert E. Stroud,Lawrence W. Dobrucki,Albert J. Sinusas +7 more
TL;DR: A proteolytically diverse portfolio exists for MT1-MMP within the myocardium and likely plays a mechanistic role in adverse LV remodeling, and a molecular mechanism for the adverse LV matrix remodeling with MT1,MMP induction is increased processing of pro-fibrotic signaling molecules.
Journal Article
Angiotensin-converting enzyme and matrix metalloproteinase inhibition with developing heart failure: Comparative effects on left ventricular function and geometry
James H. McElmurray,Rupak Mukherjee,R. Brent New,Angela C. Sampson,Mary K. King,Jennifer W. Hendrick,Aron T. Goldberg,Thomas J. Peterson,Thomas J. Peterson,Hussein Hallak,Hussein Hallak,Michael R. Zile,Francis G. Spinale +12 more
TL;DR: This study examined the effects of specific MMP inhibition, ACE inhibition, and combined treatment on LV systolic and diastolic function in a model of CHF and found a contributory downstream mechanism of ACE inhibition is an effect on MMP activity.
Journal ArticleDOI
Changes in diastolic function during development and correction of chronic LV volume overload produced by mitral regurgitation.
Michael R. Zile,Masaaki Tomita,Kazuaki Ishihara,K. Nakano,John R. Lindroth,Francis G. Spinale,M. Michael Swindle,Blase A. Carabello +7 more
TL;DR: In this article, the authors show that mitral valve replacement (MVR) causes an augmentation in left ventricular (LV) diastolic function, increasing early DV filling rate and decreasing LV stiffness.
Journal ArticleDOI
Cardiopulmonary bypass induces the synthesis and release of matrix metalloproteinases
Cassandra Joffs,Himali R. Gunasinghe,Marlina M. Multani,B.Hugh Dorman,John M. Kratz,A. Jackson Crumbley,Fred A. Crawford,Francis G. Spinale +7 more
TL;DR: Because MMPs can degrade extracellular proteins essential for maintaining normal cellular architecture and function, enhanced MMP release and activation may contribute to alterations in tissue homeostasis in the early postoperative period.
