Journal ArticleDOI
Human Xeno-autoantibodies Against a Non-human Sialic acid Serve As Novel Serum Biomarkers and Immunotherapeutics in Cancer
Vered Padler-Karavani,Nancy Hurtado-Ziola,Minya Pu,Hai Yu,Shengshu Huang,Saddam Muthana,Harshal A. Chokhawala,Hongzhi Cao,Patrick Secrest,Dinorah Friedmann-Morvinski,Oded Singer,Darius Ghaderi,Inder M. Verma,Yu Tsueng Liu,Karen Messer,Xi Chen,Ajit Varki,Richard Schwab +17 more
TL;DR: It is shown that human anti-Neu5Gc antibodies can be affinity-purified in substantial amounts from clinically approved intravenous IgG (IVIG) and used at higher concentrations to suppress growth of the same Neu 5Gc-expressing tumors.
Journal ArticleDOI
Long-Term IgG Response to Porcine Neu5Gc Antigens without Transmission of PERV in Burn Patients Treated with Porcine Skin Xenografts
Linda Scobie,Vered Padler-Karavani,Stéphanie Le Bas-Bernardet,Claire Crossan,Josef Blaha,Magda Matouskova,Ralph D. Hector,Emanuele Cozzi,Bernard Vanhove,Béatrice Charreau,Gilles Blancho,Ludovic Bourdais,Mariachiara Tallacchini,Juan M. Ribes,Hai Yu,Xi Chen,Jitka Kracikova,Broz L,Jiri Hejnar,Pavel Vesely,Pavel Vesely,Yasuhiro Takeuchi,Ajit Varki,Jean-Paul Soulillou +23 more
TL;DR: Blood of burn patients who had received living pig-skin dressings for up to 8 wk was analyzed for the presence of PERV as well as for the level and nature of their long term immune response against pig Ags, suggesting that anti-Neu5Gc Abs represent a barrier for long-term acceptance of porcine xenografts.
Journal ArticleDOI
Cross-comparison of protein recognition of sialic acid diversity on two novel sialoglycan microarrays
Vered Padler-Karavani,Xuezheng Song,Hai Yu,Nancy Hurtado-Ziola,Shengshu Huang,Saddam Muthana,Harshal A. Chokhawala,Jiansong Cheng,Andrea Verhagen,Martijn A. Langereis,Ralf Kleene,Melitta Schachner,Raoul J. de Groot,Yi Lasanajak,Haruo Matsuda,Richard Schwab,Xi Chen,David F. Smith,Richard D. Cummings,Ajit Varki +19 more
TL;DR: Two arrays that together represent the marked diversity of sialic acid modifications, linkages, and underlying glycans in nature are compared, showing that different presentations in different formats provide useful and complementary interpretations of glycan-binding protein specificity.
Journal ArticleDOI
Broad and direct interaction between TLR and Siglec families of pattern recognition receptors and its regulation by Neu1.
Guo-Yun Chen,Nicholas K. Brown,Wei Wu,Zahra Khedri,Hai Yu,Xi Chen,Diantha van de Vlekkert,Alessandra d'Azzo,Pan Zheng,Yang Liu +9 more
TL;DR: The data raised an intriguing possibility of a broad repression ofTLR function by Siglecs and a sialidase-mediated de-repression that allows positive feedback of TLR activation during infection.
Journal ArticleDOI
Pasteurella multocida sialic acid aldolase: a promising biocatalyst.
TL;DR: Substrates specificity studies showed that 5-O-methyl-ManNAc, a ManNAc derivative, can be used efficiently as a substrate by PmNanA, but not efficiently by EcNAnA, for the synthesis of 8- O-methyl Neu5Ac.
