Single-cell RNA-seq reveals new types of human blood dendritic cells, monocytes, and progenitors.
Alexandra-Chloé Villani,Alexandra-Chloé Villani,Rahul Satija,Rahul Satija,Gary Reynolds,Siranush Sarkizova,Karthik Shekhar,James Fletcher,Morgane Griesbeck,Andrew Butler,Shiwei Zheng,Suzan Lazo,Laura Jardine,David Dixon,Emily Stephenson,Emil Nilsson,Ida Grundberg,David McDonald,Andrew Filby,Weibo Li,Weibo Li,Philip L. TL;DR: This refined analysis has identified, among others, a previously unknown dendritic cell population that potently activates T cells and reclassify pDCs as the originally described “natural interferon-producing cells (IPCs)” with weaker T cell proliferation induction ability.

TL;DR: In this paper, the authors performed single-cell RNA sequencing of ~2400 cells isolated from healthy blood donors and enriched for HLA-DR + lineage − cells, which led them to identify and validate six Dendritic cells (DCs) and four monocyte subtypes.

Teichmann,Sarah A. Teichmann,Muzlifah Haniffa,Muzlifah Haniffa +62 more
TL;DR: A shift in the haemopoietic composition of fetal liver during gestation away from being predominantly erythroid, accompanied by a parallel change in differentiation potential of HSC/MPPs is demonstrated, which is validated to produce an integrated map of fetal liver haematopoiesis.
Journal ArticleDOI

TL;DR: In this paper, the transcriptomes of more than 500,000 single cells from developing human fetal skin, healthy adult skin, and adult skin with atopic dermatitis and psoriasis were compared across development, homeostasis, and disease.
Journal ArticleDOI
Gene expression variability across cells and species shapes innate immunity
Tzachi Hagai,Tzachi Hagai,Xi Chen,Ricardo J. Miragaia,Ricardo J. Miragaia,Raghd Rostom,Raghd Rostom,Tomás Gomes,Natalia Kunowska,Johan Henriksson,Jong-Eun Park,Valentina Proserpio,Giacomo Donati,Lara Bossini-Castillo,Felipe A. Teichmann,Sarah A. Teichmann,Sarah A. Teichmann +28 more
TL;DR: The innate immune response’s transcriptional divergence between species and variability in expression among cells is characterized, suggesting that this expression pattern has evolved as a mechanism for fine-tuned regulation to achieve an effective but balanced response.
