Heterozygous Deficiency of PHD2 Restores Tumor Oxygenation and Inhibits Metastasis via Endothelial Normalization
Massimiliano Mazzone,Massimiliano Mazzone,Daniela Dettori,Daniela Dettori,Daniela Dettori,Rodrigo Leite de Oliveira,Rodrigo Leite de Oliveira,Sonja Loges,Sonja Loges,Thomas Schmidt,Thomas Schmidt,Bart Jonckx,Bart Jonckx,Ya Min Tian,Anthony A. Lanahan,Patrick J. Pollard,Carmen Ruiz de Almodovar,Carmen Ruiz de Almodovar,Frederik De Smet,Frederik De Smet,Stefan Vinckier,Stefan Vinckier,Julián Aragonés,Julián Aragonés,Koen Debackere,Koen Debackere,Aernout Luttun,Sabine Wyns,Sabine Wyns,Bénédicte F. Jordan,Alberto Pisacane,Bernard Gallez,Maria Grazia Lampugnani,Elisabetta Dejana,Michael Simons,Peter J. Ratcliffe,Patrick H. Maxwell,Peter Carmeliet,Peter Carmeliet +38 more
TL;DR: In this paper, the role of endothelial PHD2 in vessel shaping by implanting tumors in mice was investigated. But, the authors did not find that PHD deficiency did not affect vessel density or lumen size.
Journal ArticleDOI
Macrophage skewing by Phd2 haplodeficiency prevents ischaemia by inducing arteriogenesis
Yukiji Takeda,Sandra Costa,Estelle Delamarre,Carmen Roncal,Rodrigo Leite de Oliveira,Mario Leonardo Squadrito,Veronica Finisguerra,Sofie Deschoemaeker,Françoise Bruyère,Mathias Wenes,Alexander Hamm,Jens Serneels,Julie Magat,Tapan Bhattacharyya,Tapan Bhattacharyya,Andrey Anisimov,Bénédicte F. Jordan,Kari Alitalo,Patrick H. Maxwell,Bernard Gallez,Zhen W. Zhuang,Yoshihiko Saito,Michael Simons,Michele De Palma,Massimiliano Mazzone +24 more
TL;DR: Unraveling how PHD2 regulates arteriogenesis and artery homeostasis by controlling a specific differentiation state in macrophages is unraveled and new treatment options for ischaemic disorders are suggested.
Journal ArticleDOI
An Acquired Vulnerability of Drug-Resistant Melanoma with Therapeutic Potential
Liqin Wang,Rodrigo Leite de Oliveira,Sanne Huijberts,Evert Bosdriesz,Nora Pencheva,Diede Brunen,Astrid Bosma,Ji-Ying Song,John Zevenhoven,G Tjitske Los-de Vries,Hugo M. Horlings,Bastiaan Nuijen,Jos H. Beijnen,Jan H.M. Schellens,René Bernards +14 more
TL;DR: In a study in patients with advanced BRAF+MEK inhibitor-resistant melanoma, vorinostat can selectively ablate drug-resistant tumor cells, providing clinical proof of concept for the novel therapy identified here.
Journal ArticleDOI
Inducing and exploiting vulnerabilities for the treatment of liver cancer.
Cun Wang,Cun Wang,Serena Vegna,Haojie Jin,Haojie Jin,Bente Benedict,Cor Lieftink,Christel Ramirez,Rodrigo Leite de Oliveira,Ben Morris,Jules Gadiot,Wei Wang,Aimee du Chatinier,Liqin Wang,Dongmei Gao,Bastiaan Evers,Guangzhi Jin,Zheng Xue,Arnout Schepers,Fleur Jochems,Antonio Mulero Sanchez,Sara Mainardi,Hein te Riele,Roderick L. Beijersbergen,Wenxin Qin,Leila Akkari,René Bernards +26 more
TL;DR: Using multiple in vivo mouse models of liver cancer, treatment with combined inhibition of of CDC7 and mTOR results in a marked reduction of tumour growth, indicating that exploiting an induced vulnerability could be an effective treatment for liver cancer.
Journal ArticleDOI
High-Throughput Functional Genetic and Compound Screens Identify Targets for Senescence Induction in Cancer
Liqin Wang,Rodrigo Leite de Oliveira,Cun Wang,João M. Fernandes Neto,Sara Mainardi,Bastiaan Evers,Cor Lieftink,Ben Morris,Fleur Jochems,Lisa Willemsen,Roderick L. Beijersbergen,René Bernards +11 more
TL;DR: A one-two punch approach for the treatment of cancer in which a drug is first used to induce senescence in cancer cells and a second drug is then used to kill senescent cancer cells is proposed.
