TL;DR: Time-resolved serial femtosecond crystallography at an x-ray free electron laser shows how motions are choreographed as bR transports protons uphill against a transmembrane concentration gradient.
Journal ArticleDOI
Membrane protein structure determination by SAD, SIR, or SIRAS phasing in serial femtosecond crystallography using an iododetergent
Takanori Nakane,Shinya Hanashima,Mamoru Suzuki,Haruka Saiki,Taichi Hayashi,Keisuke Kakinouchi,Shigeru Sugiyama,Satoshi Kawatake,Shigeru Matsuoka,Nobuaki Matsumori,Eriko Nango,Jun Kobayashi,Tatsuro Shimamura,Kanako Kimura,Chihiro Mori,Naoki Kunishima,Michihiro Sugahara,Yoko Takakyu,Shigeyuki Inoue,Tetsuya Masuda,Toshiaki Hosaka,Kensuke Tono,Yasumasa Joti,Takashi Kameshima,Takaki Hatsui,Makina Yabashi,Tsuyoshi Inoue,Osamu Nureki,So Iwata,Michio Murata,Eiichi Mizohata +30 more
TL;DR: The iodine-containing detergent provided strong anomalous and isomorphous difference signals, which enabled experimental phasing using lower-resolution reflections from fewer indexed images than phasing attempts reported previously, paving the way for de novo structure determination of membrane proteins, which often diffract poorly, even with the brightest XFEL beams.
Journal ArticleDOI
Stereoselective synthesis of the head group of archaeal phospholipid PGP-Me to investigate bacteriorhodopsin–lipid interactions
Jin Cui,Satoshi Kawatake,Yuichi Umegawa,Sébastien Lethu,Masaki Yamagami,Shigeru Matsuoka,Fuminori Sato,Nobuaki Matsumori,Michio Murata +8 more
TL;DR: The first synthesis of the bisphosphate head group of PGP-Me was carried out using H-phosphonate chemistry that led to the production of a simplified P GP-Me analogue with straight alkyl chains, suggesting that the bisPhosphate moiety plays a role in the proper functioning of bR through the lipid-protein interaction.
Journal ArticleDOI
Evaluation of diacylphospholipids as boundary lipids for bacteriorhodopsin from structural and functional aspects.
TL;DR: The present study suggests that bR reconstituted in DOPG bilayers may possibly be used as a model system for spectroscopic investigations of the lipid-bR interactions with the membrane-integral α-helices, and potentially for a similar type of membrane proteins.

Browning-reduced and enzyme-treated product of larva of bee, loyal jelly and extract thereof, and method for preparing the same
TL;DR: In this article, a method for inhibiting browning phenomena caused when a larva of a bee, loyal jelly or an extract thereof is subjected to proteolytic enzyme treatment is proposed.
