Journal ArticleDOI
Content-aware image restoration: pushing the limits of fluorescence microscopy.
Martin Weigert,Uwe Schmidt,Tobias Boothe,Andreas Müller,Alexandr Dibrov,Akanksha Jain,Benjamin Wilhelm,Deborah Schmidt,Coleman Broaddus,Siân Culley,Siân Culley,Mauricio Rocha-Martins,Fabián Segovia-Miranda,Caren Norden,Ricardo Henriques,Ricardo Henriques,Marino Zerial,Michele Solimena,Jochen C. Rink,Pavel Tomancak,Loic Royer,Florian Jug,Eugene W. Myers,Eugene W. Myers +23 more
TL;DR: This work shows how content-aware image restoration based on deep learning extends the range of biological phenomena observable by microscopy by bypassing the trade-offs between imaging speed, resolution, and maximal light exposure that limit fluorescence imaging to enable discovery.
Journal ArticleDOI
Fast live-cell conventional fluorophore nanoscopy with ImageJ through super-resolution radial fluctuations.
Nils Gustafsson,Siân Culley,George W. Ashdown,Dylan M. Owen,Pedro M. Pereira,Ricardo Henriques +5 more
TL;DR: The broad applicability of SRRF and its performance at low signal-to-noise ratios allows super-resolution using modern widefield, confocal or TIRF microscopes with illumination orders of magnitude lower than methods such as PALM, STORM or STED.

Content-Aware Image Restoration: Pushing the Limits of Fluorescence Microscopy
Martin Weigert,Uwe Schmidt,Tobias Boothe,Andreas Müller,Alexandr Dibrov,Akanksha Jain,Benjamin Wilhelm,Deborah Schmidt,Coleman Broaddus,Siân Culley,Mauricio Rocha-Martins,Fabián Segovia-Miranda,Caren Norden,Ricardo Henriques,Marino Zerial,Michele Solimena,Jochen C. Rink,Pavel Tomancak,Loic Royer,Florian Jug,Eugene W. Myers,Eugene W. Myers +21 more
TL;DR: This work shows how deep learning enables biological observations beyond the physical limitations of microscopes, and illustrates how microscopy images can be restored even if 60-fold fewer photons are used during acquisition.
Journal ArticleDOI
Quantitative mapping and minimization of super-resolution optical imaging artifacts
Siân Culley,David Albrecht,Caron Jacobs,Pedro M. Pereira,Pedro M. Pereira,Christophe Leterrier,Jason Mercer,Ricardo Henriques,Ricardo Henriques +8 more
TL;DR: NanoJ-SQUIRREL is presented, an ImageJ-based analytical approach that provides quantitative assessment of super-resolution image quality by comparing diffraction-limited images and super- resolution equivalents of the same acquisition volume, and can guide researchers in optimizing imaging parameters.
Journal ArticleDOI
SRRF: Universal live-cell super-resolution microscopy.
TL;DR: SRRF can be used to retrieve super-resolution information from most common fluorescence microscopes, and is compatible with any fluorophore, including conventional fluorescent proteins such as GFP.
